P. lima에서는 잘 알려진 독소인 dinophysistoxin과 그의 유도체 및 okadaic acid 유도체 등 4개의 화합물들 외 tetrahydropyran과 1,3,5,7-tetra(methylene) 작용기를 가진 신규 limaol 화합물 1개가 분리하였다. 이 화합물들은 모두 세포독성 활성을 가지고 있었다. 본 연구에서는 배양의 어려움 때문에 많은 연구가 이루어지고 있지 않은 해양 와편모류로부터 새로운 생리활성 화합물을 분리하였다. 이는 앞으로 해양 와편모류도 생리활성이 우수한 천연물을 제공할 수 있는 좋은 생물 소재라는 것을 보여주었다.

Marine natural products have unique skeleton chemical structures compared with the chemical structure of substances made by terrestrial organisms and show various physiological activities. In particular, secondary metabolites isolated from marine dinoflagellates have been known as very complex chemical structures, and also strong toxicity which affects not only marine ecosystem but also human health problems. This study was performed on secondary metabolites isolated from two species of marine dinoflagellates (Oxyrrhis marina and Prorocentrum lima) collected from the coastal waters of Korea (Gunsan and Geomundo, respectively). The two dinoflagellates were cultivated to a large amount in laboratory and harvested by using a continuous centrifuge system to yield a pellet unit. The obtained pellet was extracted with 50% aqueous methanol. After the application of diverse chromatography, secondary metabolites were isolated and purified by HPLC from fractions showing strong toxicity in the brine shrimp lethality test or interesting signals in the 1H NMR spectrum. Following these processes, the chemical structures of the isolated well-known toxic compounds and three new compounds were determined by various spectroscopic methods including 1D NMR spectroscopy(1H and 13C), 2D NMR spectroscopy(HSQC, HMBC, COSY, TOCSY, and ROESY), along with mass spectrometry. From O. marina were isolated a new trioxyline derived from DHA and a new sulfoquinovosyl diacylglycerol (SQDG), in which unsaturated fatty acid, saturated fatty acid and sulfoquinovose are bound. The two compounds showed NO(Nitric Oxide) inhibitory effect on RAW264.7 macrophage cells. On the other hand, P. lima allowed us to isolate four compounds: dinophysistoxin (DTX), its derivatives and okadaic acid derivatives, which are well-known toxins, and also a new compound, limaol, bearing tetrahydropyran and 1,3,5,7-tetra(methylene) a functional group. All these compounds exhibited cytotoxic activity. In this study, we successfully isolated new physiologically active compounds from marine dinoflagellates which have not widely been studied because of much difficulty in culture. This study shows that marine dinoflagellates can be a good biomaterial to provide natural products with excellent physiological activity.
