Neurodegenerative disease are caused by the progressive loss of structure or function of neurons in the brain and spinal cord. Unfortunately, so far there has been no successful treatment for neurodegenerative diseases. Cell replacement therapy is considered as potentially powerful new therapeutic strategies for human neurological diseases. In recent years, neurons and glial cells have successfully been generated from stem cells, and extensive efforts by investigators to develop stem cell-based brain transplantation therapies have been carried out. In this thesis, I focused on the therapeutic effect of stem cell transplantation in animal models of Parkinson’s disease (PD) and Huntington’s disease (HD). Two human stem cell lines (H9 and ReNcell VM) were used as cell sources for PD. These cells were transplanted into 6-OHDA lesioned rat model of PD. Various stem cell lines including human neural stem cell line (HB1.F3.BDNF), human induced pluripotent stem cell derived from patient’s somatic cells (HD1 iPS) and protein-based human iPS cell line (Pro-1) were used as cell sources for HD. These cell were grafted intracerebrally in the quinolinic acid (QA)-induced rat model of HD. I studied the regenerative capacity of injured brain tissue and functional recovery using behavior tests that include stepping test, rotarod test, staircase test and drug-induced rotation test following transplantation. To track the fates of transplanted cells in vivo, I employed 4.7T animal MRI, which visualized the presence and the migration of contra-laterally transplanted cells. Collectively, these results indicate that transplantation of stem cells can replace lost partially neurons, reconstitute damaged neuronal connections, enhance endogenous neurogenesis, reduce inflammatory response, and improve behavior deficits in neurodegenerative animal models.
