As studies have reported on the dangers of mold, the importance of mold management has been emphasized. Since 2017, the recommanded concentration level for total indoor air fungi for domestic multi-use sensitive facilities has been established. However, the recommendation criteria for harmful pathogens has not yet been established. Pathogenic fungal spores can cause respiratory and skin-related diseases, and various toxins and metabolites are known to negatively influence human function. For the regulation and management of pathogenic fungi, identification of harmful species through identification and quantitative analysis of concentration is necessary. However, no standard operation procedure (SOP) for molds has been created yet, and hence, so it is proceeding with reference of bacteria SOP. Therefore, in this study, we tried to develop the analysis and quantification technique for harmful fungal species. First, identification analysis was performed using differences in the length of specific nucleotide sequences. The internal transcribed spacer (ITS) region was amplified by conventional polymerase chain reaction (PCR) and used for identification of the specific nucleotide sequence. We analyzed the length of the amplified ITS using a microchip-based capillary gel electrophoresis method, which has better resolution and reproducibility than the conventional flat electrophoresis method. This confirmed the differences at the genus level and helped in identification. Secondly, identification / quantitative analysis was performed by measuring the antigen-antibody reaction between spores and fungus-specific antibodies using biolayer interferometry (BLI). The cross-reactivity of the antibodies and the selected six harmful species were confirmed. A detection limit of 8.4 x 103 spores/mL was obtained via this method. In addition, signal amplification was performed by introducing an antibody conjugated with alkaline phosphatase, and a detection limit of 3.3 x 103 spores/mL was obtained.
