Plackett-Burman design에서 yeast extract, peptone, NaCl, Na2HPO4 그리고 glucose와 같은 5가지 성분들이 HA 생산에 어떤 영향을 주는지 평가 되었다. Glucose, NaCl, yeast extract는 HA 생산에 중요한 영향을 주는 것을 발견하였고, 이어서 Box-Behnken design을 이용한 반응 표면 방법론 (response surface methodology)을 통한 3개 성분의 최적 농도를 구하였다. HA 최대 생산을 위하여 Brain Heart Infusion (BHI) 배지를 기준으로 한 최적 배지 성분은 아래와 같다: glucose 21.0 g/L, NaCl 9.0 g/L, yeast extract 18.386 g/L, peptone 10.0 g/L, Na2PO4 2.5 g/L. 위 성분들은 기존 배지와 비교하여 glucose는 42.0 g/L에서 21.0 g/L로 변하였고, NaCl은 5.0 g/L에서 9.0 g/L로, 질소원은 calf brains 7.7 g/L과 beef heart 9.8 g/L (총 17.5 g/L)에서 yeast extract 18.386 g/L로 변경 하였다. 위 조건 아래에서, 최종 HA 농도는 3.71 g/L였고, 이는 종전 배지 (2.50 g/L) 48 시간 때보다 1.48배 높아진 값이다. 본 실험을 통하여 배지 조성물의 몇 가지 변화에 따라서 히알루론산의 생산량 증가를 보여줬으며, 결론적으로 이번 연구를 통해서 NaCl이 히알루론산 생산에 가장 중요한 배지 조성임을 알 수 있으며 추가적으로 농도를 조절함에 따라 더욱 좋은 배지 최적화가 가능할 것으로 보인다. 본 실험을 통한 최적 배지는 BHI와 같은 상업용 배지보다 낮은 비용으로 생산케 함 외에도, 히알루론산 생산양은 1.48 배 증가 했다. 일부 프로세스 및 배지 요소의 추가 최적화를 연구가 필요하지만, 본 연구는 C. glutamicum을 통한 히알루론산 생산성 향상을 위한 추가 실험에 대한 근거가 될 수 있다.

In this study, media optimization was conducted to figure out what conditions which are temperature, pH and media components for improved Hyaluronic acid (under HA) production from genetically engineered Corynebacterium glutamicum by Plackett-Burman design. Glucose, NaCl, yeast extract were found to have significant influence on HA production and the optimal concentrations of these 3 factors were sequentially investigated by the response surface methodology using a Box-Behnken design. The optimal medium components obtained for maximum HA production based on Brain Heart Infusion (BHI) broth with 20 % Glucose were as follows: Glucose 21.0 g/L, NaCl 9.0 g/L, Yeast extract 18.386 g/L, Peptone 10.0 g/L, Na2HPO4 2.5 g/L. These components that differ from a typical broth were Glucose changed from 42.0 g/L to 21.0 g/L, NaCl changed 5.0 g/L to 9.0 g/L, and nitrogen sources changed from Calf brains 7.7 g/L and Beef heart 9.8 g/L (total 17.5 g/L) to Yeast extract 18.386 g/L. Under these conditions, the final HA concentration was 3.71 g/L, which is about 1.48-fold higher than the original medium (2.50 g/L) at 48 h. This report showed the improvement of HA titer by some changes of media components. This research is the first systematic optimization for a HA production medium and it showed the effect of each component and environmental conditions. From those results, medium for HA production supplied cheaper than commercial medium such as BHI by replacing high cost medium component (Calf brans infusion and Beef heart infusion) and reduce surplus substrate concentration (Glucose). Moreover, comparing with previously studied media productivity of HA increased by 1.48-fold. Although some process and component optimizations are need to be studied, it gave some evidences that further experiments to enhance HA productivity in C. glutamicum.
