HaCaT 세포에 과산화수소를 1시간 처리하고 홉추출물을 4시간, 24시간, 48시간 처리한 후, 산화적 스트레스에 대한 세포 보호 효과를 측정한 결과, 25 μg/mL 농도에서 모두 우수한 세포 보호효과를 확인 하였으며, 양성 대조군으로 ascorbic acid를 사용하여 세포 내 ROS 생성 억제효과를 측정한 결과, 50μg/mL 농도에서 유의한 활성억제를 확인하였다. 홉 추출물의 항노화와 관련한 MMP-1 발현에 미치는 영향을 확인하기 위해 HDF 세포에 UVB를 사용하여 활성을 유도한 뒤 홉 추출물을 농도 별로 처리한 결과, MMP-1 활성 억제 효과는 없는 것으로 확인되었고, 또한 Ascorbic Acid를 양성 대조군으로 사용하여 홉추출물이 콜라겐 생성에는 크게 영향을 미치지 않는 것으로 확인되어, 홉추출물이 항노화에는 효과가 없는 것으로 사료된다. 이와 같은 결과로, 홉 추출물이 강력한 항산화 작용과 탁월한 세포 내 보호 효과를 가지며 티로시나제와 멜라닌의 생합성을 억제한다는 것을 확인하였고, 이는 홉추출물의 기능성 미백 화장품 원료로서의 가능성을 보여준다 할 수 있다.

In recent years, there has been a growing interest in functional cosmetic raw materials that are harmless to humans, but research on the feasibility of hops as cosmetic raw materials has been relatively inactive. In current study, Biological properties of hop extract were investigated to evaluate their applicability as functional cosmetic raw materials. Anti-oxidative effects of hop extract were investigated to determine polyphenols and flavonoids, known as antioxidants. When total polyphenol and flavonoid were measured by using caffeic acid and quercetin as standard materials, they were increased in a dose-dependent manner at all concentrations. Especially, at the concentration of 5 mg/mL of hops extract, high polyphenol content of 183 mg/g was obtained. In addition, 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging activity and the melanin biosynthesis inhibitory effect on B16F10 melanoma cells were examined. As a result, 84 % of DPPH radical scavenging activity was confirmed at a concentration of 10 μg/mL of hop extract. After inducing the production of melanin by α-MSH, the cells were treated with the hop extract, and a significant inhibitory effect on melanin production was confirmed. Also, significant inhibitory effect of MITF, Tyrosinase was determined by western blot analysis. At a concentration of 100 μg/mL of hop extract, higher inhibitory effect of M-Tyrosinase (15 %) than arbutin (6 %), a positive control, was observed. When the hop extract was treated at intervals of 10 minutes, a high inhibitory effect of C-Tyrosinase was observed after 90 minutes at a concentration of 100 μg/mL. As a result, high polyphenol contents, DPPH radical scavenging activity, and excellent inhibitory effects of melanin biosynthesis, MITF, M-Tyrosinase and C-Tyrosinase are thought to affect the anti-oxidative activity and whitening effect of hop extract. To confirm the anti-inflammatory effect of hop extract, its inhibition of NO production was measured using lipopolysaccharide-stimulated RAW 264.7 cells, and the significant inhibitory effect was demonstrated. To examine the cytoprotective effect of hop extract against oxidative stress, HaCaT cells were treated with hydrogen peroxide for 1 hour and treated with hop extract for 4 hours, 24 hours and 48 hours again. As a result, the cytoprotective effect was confirmed at 25 μg/mL. Ascorbic acid was used as a positive control to measure the inhibitory effect of intracellular ROS production. As a result, significant inhibitory effect was observed at a concentration of 50 μg/mL. In order to examine the inhibitory effect of hop extract on the expression of MMP-1, which is related to anti-aging activity, UVB was used to induce activity of HDF cells, it was confirmed that there was no significant inhibitory effect on MMP-1 activity. In addition, collagen production effect of hop extract was measured using ascorbic acid as a positive control, and confirmed that there was no significant effect of collagen production as well. It suggests that hop extract is not effective for anti - aging. As a result, it was confirmed that the hop extract had a strong antioxidative effect and an excellent cytoprotective effect and inhibited the biosynthesis of tyrosinase and melanin. This shows that the hop extract can be used as a functional whitening cosmetic raw material.
