통증은 실재적 혹은 잠재적 조직 손상에 의해 야기되는 불쾌한 감각 및 정서적 경험을 말하며, 통각수용기와 신경섬유로 구성된 구심성 신경로를 통하여 일어나는 통각 및 감각 장애를 뜻하기도 한다. 우리 몸의 여러 기관들에 유해성 자극이 오게 되면, 일차감각뉴런의 자유 신경 말단에 있는 통각수용기를 탈분극 시키고, 이 신호가 신경 섬유를 통해 척수에 도달하게 된다. 그러면 섬유 말단의 뉴런에서 글루탐산염과 신경펩타이드 같은 신경 전달 물질이 방출 되어 N-메틸-D-아스파트산염 (NMDA) 수용체를 포함한 척수배각뉴런의 수용체를 활성화시키는 과정으로 일어나게 된다. 하지만, 반복적으로 유해 자극에 노출될 경우 통각수용기의 민감화가 발생하여 통각 과민을 초래할 수 있고 더 나아가 척수배각뉴런 흥분성의 장기적 변화를 일으키는 데 이러한 통증 반응 과정에서 NMDA 수용체가 중요하게 관여하는 것으로 보고 되어 있다. 따라서, 본 논문에서는 쥐에서 통증/반복 행동에 대한 NMDA 복강 투여의 효과를 확인하고 그에 따른 NMDA 수용체와 통증 자극 기전 (ERK, CamKⅡ)의 발현을 확인하기 위하여 실시하였다. 본 실험은 ICR 수컷 쥐를 사용하였고, 모든 주사 방법은 복강 투여로 실시하였다. 대부분의 연구는 그루밍과 꼬리를 물거나 핥는 행동을 측정하기 위해 행동 관찰을 실시하였으며, 통증과 관련된 반응을 알기 위해 hot plate test와 writhing test를 진행하였고, NMDA 수용체 소단위와 통증 자극 기전 (ERK, CamKⅡ)의 발현을 확인하기 위해 western blot을 실시하였다. 쥐에 똑같은 농도의 NMDA를 복강 투여하였을 때, 그루밍과 꼬리를 물거나 핥는 통증/반복 행동이 관찰되었다. 그래서, 그루밍과 꼬리를 물거나 핥는 행동의 시간이나 횟수를 각각 나누어 측정하였다. 그 결과, NMDA 복강 투여는 꼬리를 물거나 핥는 행동을 증가시켰고, NMDA의 농도가 높고 쥐가 어릴수록 꼬리를 물거나 핥는 행동이 증가되는 것을 확인하였다. 또한, hot plate test에서 NMDA를 투여한 군, 특히 75 mg/kg군에서 발을 핥거나 뛰어오르는 데까지 걸리는 잠재 시간이 줄어드는 것을 확인하였다. S + A군과 NMDA를 투여한 군을 비교해 보았을 때, 몸부림 치는 횟수가 증가하였지만, 유의적인 차이는 없었다. 흥미롭게도, 투여하고 24시간 뒤 NMDA를 투여한 군의 몇몇 쥐들이 그들의 꼬리를 물어 상처가 생긴 것을 확인하였고, 그 상처의 정도는 각자 달랐다. 또한, 몇몇 쥐들은 꼬리를 무는 것으로부터 보이는 상처가 보이지 않았다. 마지막으로, 비록 NMDA 수용체와 통증 자극 기전 (ERK, CamKⅡ)의 발현이 크게 차이가 나진 않지만, NMDA 투여에 의해 척수의 NMDA 수용체 발현이 증가하는 것으로 보아 NMDA 복강 투여는 척수의 NMDA 수용체, 특히 NR2A의 발현의 증가로 통증 행동을 증가시키는 것으로 추측된다. 본 논문의 흥미로운 점은 1) NMDA 복강 투여가 꼬리를 물거나 핥는 행동을 증가시킴 2) 그 효과는 주령 의존적으로 나타남 3) 같은 농도의 NMDA를 투여하여도 그룹간의 차이가 있음 이다. 이러한 결과는 추가 연구를 통해 본 연구가 앞으로의 통증 연구 및 치료제 개발에 기여할 것으로 사료된다.

Pain is unpleasant sensory and emotional experiences caused by actual or potential tissue damage and also refers to nociceptive and sensory disturbances that occur through the afferent nervous system composed of nociceptors and nerve fibers. When a harmful stimulus comes to the various organ of our body, depolarizes the nociceptors at the free nerve ending of primary sensory neurons, and the signal reaches the spinal cord through the nerve fibers. Then, neurotransmitters such as glutamate and neuropeptides from the neurons of nerve fibers are released to activate receptor of the dorsal horn neurons including N-methyl-D-aspartate (NMDA) receptor. However, when repeatedly exposed the repetitive noxious stimuli, sensitization of the nociceptors may occur, resulting in hyperalgesia and furthermore, a long-term change in the excitability of dorsal horn neurons. In such a nociceptive reaction, NMDA receptors have been reported to play an important role. Therefore, this study was conducted to confirm the effects of intraperitoneal NMDA administration on nociceptive/repetitive behaviors in mice and to identify the expression of NMDA receptor subunit and nociceptive sensation pathway (ERK, CamKⅡ). We tested using ICR male mice, and all methods of injection were intraperitoneal administration. And, most of our studies proceeded with behavioral observations for measurement of grooming and biting, licking behaviors, hot plate test, writhing test to know the pain-related reactions, and western blot analysis to identify expression of NMDA receptor subunit and nociceptive sensation pathway (ERK, CamKⅡ). When the mice were injected intraperitoneal NMDA administration, nociceptive and repetitive behaviors were observed like grooming and tail biting, licking behaviors. So, the duration and the number of grooming or tail biting, licking behaviors were measured separately. As a result, we found that the intraperitoneal NMDA administration increased the tail biting, licking behaviors and that higher dosage of NMDA enhanced the tail biting, licking behaviors, especially in younger mice. Also, in hot plate test, we found that the latency of paw licking and jumping was decreased in the NMDA-exposed mice especially at 75 mg/kg group. The number of writhing was enhanced in NMDA-exposed groups compared with the S + A group, but there were no statistical differences. Interestingly, we observed that the some NMDA injected mice bit their tail a lot, resulting in injury on the mice’s tail after 24 h of injection but the degree of injury was also different individually. Some mice in the NMDA-injected group had no manifestations of wound from tail biting. Finally, although the expression of NMDA receptor and the nociceptive sensation pathway (ERK, CamKⅡ) were not significantly different, an increased expression of NMDA receptor in the spinal cord, particularly NR2A, was observed after intraperitoneal NMDA administration, which could explain the increased nociceptive behaviors. New findings in my research included that 1) intraperitoneal NMDA administration induced tail biting, licking behaviors 2) effects of NMDA by age-dependent, and 3) Group variance in NMDA-exposed mice at same concentration. These results are expected to contribute to the development of future pain research and treatments through further studies.
