TCS7010은 ROS의 생성을 유도하여, UPR 경로를 자극하고 CHOP 발현을 증가시켰으며, 그 하위 기질인 Bim의 발현 또한 증가하였다. Deregulation of apoptosis can disrupt the balance between cell death and proliferation, and cause the disease like cancer (Danial, 2004). The inability that respond to death sign such as cellular stress, harmful mutations and DNA damage results in the uncontrolled growth of the tumor (Thompson, 1995). Imbalance with cellular system make cancer cells more sensitive to stress. The endoplasmic reticulum (ER) plays a crucial role in protein maturation. Accumulation of misfolded protein provokes ER stress condition and activates ER stress sensors; PERK, IRE1, and ATF6 (Kaufman, 1999). Activated sensors trigger a series of events known as the unfolded protein response (UPR) to restore homeostasis, which serves to enhance protein folding ability, and reduce protein synthesis. The major factor inducing ER stress is reactive oxygen species (ROS), which contain oxygen, including singlet oxygen, peroxides (H2O2), superoxide (•O2−), and hydroxyl radical (•OH). ROS are formed as a natural byproduct of the normal cellular metabolism of oxygen, and they are removed by own redox system. However environmental stress, such as UV, can increase ROS level dramatically. It may result in damage to ER by modification of protein and lipid structure. Oxidative stress triggers ER stress. If ER stress continued over the threshold, it causes UPR to induce apoptosis pathway (A. Bartolotti, 2000). This study investigated the molecular mechanism of inducing apoptosis through ROS-mediated ER stress by synthetic chalcone derivatives and TCS7010 in cancer cells. In chapter 1, to find more effective chemopreventive agents, 24 synthetic flavonoids derivatives were synthesized using DPP-23 (S.Y Shin, 2014) as a backbone structure. One of them, 2'-Hydroxy-2,3,5'-trimethoxychalcone (DK143), induced apoptosis through the activation of caspase signaling in MDA-MB-231 breast cancer cells, whereas there is no effects on viability of MCF12A non-transformed breast epithelial cells. Furthermore, only in MDA231 cells, not MCF12A, DK143 provoked reactive oxygen species (ROS). Also expression of ER-chaperone GRP78/Bip, and ER-stress sensors were upregulated by DK143. In addition, XBP-1 and CHOP, known as UPR-related transcription factors, were activated. Moreover, transient knockdown of IRE1α or CHOP by corresponding siRNA molecules abrogated DK143-induced apoptosis. Furthermore, DK143 suppressed mouse tumor growth in vivo. Aurora kinase A (AURKA) is essential for regulating mitosis and is frequently amplified in various cancer cell types. But it remains unclear how AURKA inhibition effects on the induction of apoptosis. In chapter 2, it was shown that treatment with TCS7010, a specific inhibitor of AURKA, caused cell cycle arrest in G2/M phase, followed by accumulation of cells in the sub-G0 phase of the cell cycle and increased the percentage of annexin V-binding cells. The cleaved caspase-2, caspase-7 and PARP significantly increased following TCS7010 treatment. In addition, TCS7010 resulted in the production of reactive oxygen species (ROS). Then it stimulated UPR pathway, leading to the upregulation of CHOP, and its downstream target Bim. Pretreatment with NAC, a ROS scavenger, significantly attenuated TCS7010-induced accumulation of CHOP, Bim, cleaved caspase-7, and cleaved PARP. In summary, these results suggest that DK143 and TCS7010 induce apoptosis through promoting ROS-mediated ER stress in breast, or colon cancer cells. It might be a promising strategy for finding new chemotherapeutic or chemopreventive agents for cancer.
