Next generation sequencing을 이용한 군집분석을 통해 주요한 남조류 뿐만 아니라 Planktothrix, Lyngbya, Scytonema와 같이 10% 미만의 rare taxa까지도 분석할 수 있었다. 뿐만 아니라 형태적으로 유사하여 현미경 관찰로 구분하기 어려운 Chrococcale 목에 포함된 종들도 동정할 수 있었다. 또한 유전자 primer를 이용하여 rare taxa 세포에 존재하는 염기서열의 계통학적 차이를 통해 북한강 수계에서 Scytonema sp.가 saxitoxin 생성 잠재성을 보유하는 것으로 확인하였다. 유해 남조류의 2-MIB (2-methylisoborneol) 생성 잠재성을 판단하기 위해 제작한 유전자 primer는 실내분석에서 2-MIB를 생성하는 Pseudanabaena galeata 만을 효과적으로 탐색하였을 뿐만 아니라 의암호 수역의 공지천 퇴적물에서도 남조류의 2-MIB 형성 유전자를 탐색할 수 있었다. 염기서열을 계통학적으로 분석한 결과, Pseudanabaena 속 (genus) 남조류 세포에 존재하는 2-MIB 생성 유전자로 판단되었으며, 조사기간 동안 공지천 퇴적물에서 2-MIB를 생성할 수 있는 Pseudanabaena 속 남조류 세포밀도를 100∼100,000 cells/mL 범위로 추정하였다. 유전자 primer를 사용하여 유해 남조류 세포의 거동을 분석한 결과, 북한강 수계의 수층과 퇴적층에서 microcystin, geosmin, 2-MIB를 생성하는 유해 남조류는 의암호 수역 퇴적층에서 집중적으로 분포하였으며 특히 공지천 퇴적층에서 주로 나타났다. 퇴적층에서 수체로 유해 남조류 세포의 이동은 2월 ~ 3월에 발생하였으며 대발생이 감소하는 9월 ~ 12월에 침강하여 퇴적층에서 seed bank를 형성하였다. 유해물질 생성 유전자는 우점한 남조류 (Microcystis aeruginosa, Dolichospermum circinale, Pseudanabaena limnetica) 세포 밀도와 유의한 상관관계를 나타내었으며 (r>0.69, p<0.01), 특히 gys1 유전자와 D. circinale 세포 밀도 사이에서 상관성이 가장 높았다 (r=0.858, p<0.01). 북한강 수계에서 우점하는 M. aeruginosa와 D. circinale (straight type) 세포에서 각각 유전자 수준에서 microcystin과 geosmin 생성 잠재성이 발견되었으며, D. circinale (straight type)세포는 geosmin과 microcystin 잠재성을 모두 나타내었다. 이밖에도 Aphanizomenon flos-aquae 세포에서 microcystin 잠재성이 발견되었으며, Oscillaotria limosa 세포에서 2-MIB 생성 잠재성이 발견되었다. 북한강 수계에서 분리된 4개 유해 남조류 strain들은 공통적으로 최적 성장 온도 (25℃)에서 유해물질의 생산력 (Chl-a당 유해물질 농도)이 감소하였으며, 저온 (15℃)에서는 성장률은 낮았지만 유해물질의 생산력이 증가하였다. M. aeruginosa, D. circinale, P. galeata 에서 유해물질 생성 유전자 RNA 발현량은 유해물질 농도와 높은 상관관계를 나타내었으며 (r>0.80, p<0.01), 이를 통해 산정된 회귀식을 현장에 적용하였다. 회귀식과 변환상수를 통해 계산된 유해물질 농도는 실측된 유해물질 농도와 20∼30% 차이가 있었으나 두 요인 사이의 상관관계는 0.0001 수준에서 통계적으로 매우 유의 하였다 (R=0.854). 결론적으로 분자유전학적 기법은 전체밀도의 10% 미만으로 발생하는 rare species까지 효과적으로 탐색할 수 있었으며, 현장의 매우 낮은 밀도 수준에서도 남조류의 유해물질 생성 잠재성을 판단할 수 있었다. 본 연구에서 제작된 2-MIB primer는 남조류가 대발생하는 지역에서 2-MIB 생성 잠재성을 효과적으로 파악하여 이취미 형성 남조류 발생을 조기에 예측할 수 있다고 판단된다. 또한 남조류의 유해물질 생성 유전자를 탐색하는 primer는 남조류 세포 관찰이 어려운 퇴적물에서 유해 남조류의 발생과 유해물질 형성을 파악할 수 있으며, 이를 통해 퇴적층에서 seed bank의 거동과 유해물질 발생을 사전에 파악할 수 있다. 본 연구의 결과는 북한강 수계에서 발생하는 남조류에 의한 유해물질 발생 잠재성에 대한 유용한 자료를 제공할 뿐만 아니라, 다양한 담수생태계에서 유해 물질을 생성하는 남조류들에 대한 효과적인 탐색과 함께 남조류의 발생 잠재성 및 거동을 이해하는데 효과적인 도구로 활용이 기대된다.

This molecular genetic study evaluated the harmful materials producing potential of cyanobacteria in the North Han River. Toward this end, the next generation sequencing (NGS: shotgun metagenome sequencing) was used to analyze the diversity of cyanobacterial communities in the Sambong-ri site. Gene primers were used to determine the harmful materials producing potential and the dynamics of harmful cyanobacteria in the community. In addition, we analyzed the cyanobacterial gene expression underlying the harmful materials producing genes to predict the harmful level generated. Next generation sequencing facilitated the analysis of not only the major dominant species but also the rare taxa (10%) such as Planktothrix, Lyngbya, and Scytonema. Further, the species included in the order Chroococcale, which are morphologically similar and difficult to distinguish microscopically, were identified. The gene primers were used to detect the cellular potential for saxitoxin production by the rare taxa. Based on the phylogenic differences of the nucleotide sequence, we confirmed that Scytonema sp. has the potential to produce saxitoxin in the North Han River. Genetic primers designed to determine the potential for 2-MIB (2-methylisoborneol) synthesis by toxic cyanobacteria, effectively identified not only Pseudanabaena galeata cells in situ, but also detected the 2-MIB-producing gene in the cyanobacteria present in the Kong-ji Stream sediment of the Uiam Lake. The results of the phylogenic analysis indicated a 2-MIB-producing gene in the cells of the genus Pseudanabaena. Investigations revealed that the Kong-ji Stream sediment carried the genus Pseudanabaena at an estimated density of 100〜100,000 cells/mL, capable of producing 2-MIB. Analysis of the dynamics of the toxic cyanobacterial cell density using gene primers revealed that harmful cyanobacteria producing microcystin, geosmin, and 2-MIB in the North Han River are concentrated in the sediment of the Uiam Lake, especially in the Kong-ji Stream. The recruitment of cyanobacterial cells from the sediment to the water column occurred from February to March and precipitated from September to December when the bloom decreased, forming a seed bank. The harmful materials producing genes showed a high correlation with cell density of the dominant cyanobacteria (M. aeruginosa, D. circinale, P. limnetica) (r> 0.69, p <0.01), and in particular, the correlation between gys1 gene and D. circinale cell density was the highest (r = 0.858, p 0.80, p <0.01), and the calculated coefficients of regression were applied to the field. The regression equation and the conversion constant were used to determine the harmful material concentrations, which varied from the measured concentrations by 20-30%, however, the correlation between the two factors was highly significant at 0.001 level (r = 0.823). In conclusion, the molecular genetic method effectively enabled the detection of rare species in the cyanobacterial assemblages, and the potential for the production of cyanobacterial toxins was determined at a very low density. The 2-MIB primer designed in this study can be used to predict the potential for 2-MIB production in the region containing the cyanobacteria, and to detect the presence of odor-forming cyanobacteria. In addition, a primer for the identification of harmful materials producing gene expression in cyanobacteria can be used to detect the presence of harmful cyanobacteria and harmful materials accumulation in the sediment. Thus, the dynamics of the seed bank and the presence of harmful materials of cyanobacterial origin can be identified in advance. The present study demonstrates the role of molecular genetics in understanding the behavior of cyanobacterial species and the potential for its toxicity in the freshwater system.
