Skin whitening agents known to date often directly inhibit tyrosinase activity in epidermal melanocytes and this may result in vitiligo as its side effect. Then we are looking for novel, effective natural ingredients of absent side effects. In this study, the whitening effect and its molecular mechanisms on melanogenesis of fermented licorice extracts were investigated by In vitro. The change of major active ingredients in the licorice water extracts(LWE) and fermented licorice water extracts(FLWE) were also confirmed by TLC and HPLC. Licorice was fermented microbes of I. orientalis MFST. FLWE significantly reduced α-MSH(α-melanocyte stimulating hormone) stimulated tyrosinase activity and melanin contents without cytotoxicity in mouse melanoma B16F10 cell. In molecular mechanisms study on melanogenesis, FLWE was effectively downregulated MITF, tyrosinase, tyrosinase-related protein (TRP)-1 and TRP-2 which are key transcription factor for tyrosinase expression in pigmentation. And FLWE showed no tyrosinase inhibiting activity which causes side effects such as vitiligo. Analysis of major active ingredients in the FLWE by TLC and HPLC showed that melanogenesis related component are increased such as Isoliquiritigenin, Glycyrrhizic acid, Licochalcone A and Glabridin. These results suggest that glycosides in licorice major active molecules were converted into non-saccharide(aglycone) by fermentation. Many studies were carried out to investigate changes of biologically active components in licorice by fermentation and interest in this is increasing. Especially, in this study was found that fermentation of licorice with I. orientalis MFST increased the major components effective for whitening.
