강황 (Curcuma longa L.) 은 식품의 성분에서 의약품에 이르기까지 다양한 목적으로 사용되어 왔으며, 활성성분인 커큐민 (curcumin) 은 폭넓게 생리활성에 대한 연구가 진행되어 왔다. 그러나, 커큐민이 많이 포함된 강황 추출물은 상대적으로 덜 주목 받았으며 간 이상 지질 혈증의 치료제로서의 가능성과 그 작용기전에 대해서는 연구가 적었다. 본 연구에서는 강황 추출물과 커큐민이 쥐 모델에서의 사염화탄소 (CCl4) 에 의한 손상으로부터 간을 보호하는 메커니즘을 조사 하였다. 강황 추출물과 커큐민은 산화 스트레스를 경감시키고 지질 과산화를 억제하며 glutathione peroxidase 활성화를 증가시킴으로써 간을 CCl4에 의한 손상으로부터 보호하였다. 강황 추출물과 커큐민은 혈청 AST 와 ALT의 활성을 감소시키고 간에서의 글루타티온 함량을 향상시킴으로써 간 손상을 보호하였다. 관련된 기전 연구로, 급성 및 만성 스트레스에 대한 강황 추출물 및 커큐민의 효과에 대한 ROS를 중점적으로 연구하였다. 만성 모델에서 축적 된 ER 유래의 ROS는 급성과 차이가 있었다. 특히, 만성 스트레스에서 ER 스트레스는 지속적으로 유도되었으며, 대표적으로 PDI의 산화적 ER 단백질 접힘 상태가 손상되었다. 강황 추출물과 커큐민은 만성 스트레스 상태에서 PDI와 apoB가 결합하여 변형 된 ER 접힘 상태를 회복시키고, ER 스트레스와 결과적으로 간에서의 이상 지질 혈증을 조절 하였다. 이상의 연구는 강황 추출물과 커큐민의 ER 스트레스 조절이 지질대사 장애를 완화시키는 주요한 메커니즘임을 시사한다. 그럼에도 불구하고, 강황 추출물과 커큐민은 낮은 용해성과 그로 인한 낮은 생물학적 활용성을 갖고 있다. 따라서, 강황 추출물을 캡슐화 하기 위한 나노에멀젼의 개발이 필요하였다. 강황 추출물을 나노에멀젼화 하여 햄에 적용하였을 때 관능적으로 차이가 적음을 확인하였다. 비록 햄 속에 함유된 커큐미노이드의 양은 적지만 담체가 용해성을 증가시켜 건강에 도움을 줄 수 있을 것이다. 본 연구에서는 강황 추출물과 커큐민이 스트레스 조건에서 CCl4에 의한 간 독성으로부터의 회복을 촉진 시키며, 간 활성을 회복시킬 수 있는 치료제가 될 수 있음을 밝혔다. 또한, 강황 추출물 나노에멀젼은 기능성 식품 소재로 활용될 수 있음을 제시하였다.

Turmeric (Curcuma longa L.) has been used for many purposes from food ingredient to medicine and its active compound curcumin has been studied a wide spectrum of biological activity. Although the curcumin have been extensively studied, curcumin-rich turmeric extract received less attention, and known little about their therapeutic potential against hepatic dyslipidemia and mode of action. In this study, the mechanism by which turmeric extract and curcumin protect the liver against carbon tetrachloride (CCl4)-induced injury in rats were investigated. Turmeric extract and curcumin protect the liver from CCl4-induced liver injury by reducing oxidative stress, inhibiting lipid peroxidation, and increasing glutathione peroxidase activation. Turmeric extract and curcumin significantly protect against liver injury by decreasing the activities of serum aspartate aminotransferase and alanine aminotransferase and by improving the hepatic glutathione content. In addition, the effects of turmeric extract and curcumin against acute and chronic stress were investigated, particulary focusing on ROS. ER-originated ROS accumulated in the chronic model, another difference between the two conditions. ER stress was induced consistently, and oxidative intra-ER protein folding status, representatively PDI, was impaired, especially in chronic stress. The PDI-associated client protein hepatic apoB accumulated with the PDI-binding status in chronic stress, and curcumin recovered the altered ER folding status, regulating ER stress and the resultant hepatic dyslipidemia. Taken together, this study demonstartes that the regulation of ER stress is the main mechanism by which turmeric extract and curcumin counteract lipid dysmetabolism. Nevertheless, turmeric extract and curcumin have poor water solubility and low bioavailability. So, it is necessary to develop a nanoemulsion formulation for encapsulating turmeric extract. Turmeric extract nanoemulsions were successfully incorporated into the canned hams, less affecting their sensory characteristics. Even though the amount of nanoencapsulated curcuminoids in the ham is low, the carrier increases their solubility so they could still provide health benefits. In summary, turmeric extract and curcumin promoted recovery from CCl4-induced hepatic toxicity in both stress conditions and might be possible therapeutic agents to recover liver activity. And turmeric extract nanoemulsions were developed and can be used as a functional food ingredient.
