본 연구에서, 우리는 바이오 물질인 커큐민이 포함된 양친성 고분자 형광 코어-쉘 나노입자를 제조하였다. 형광 고분자입자는 친수성 폴리에틸렌이민 (PEI) 쉘과 소수성 폴리메틸메타크릴레이트 (PMMA) 코어 그리고 형광물질 커큐민이 코어의 PMMA에 적재되어 구성되었다. 나노입자는 아미노기를 포함하는 PEI와 MMA의 계면활성제를 사용하지 않는 그라프트 공중합법을 통해 간단한 중합과정과 정제과정으로 준비되었다. 제조된 나노입자는 모노머 농도와 반응 시간 조절을 통한 입자크기 조절이 자유롭다는 특징을 가진다. 입자크기의 변화는 제타 입도 분석기를 통해 확인하였다. 낮은 모노머의 농도와 짧은 반응 시간은 입자크기를 감소 시켰고 증가된 모노머 농도와 반응 시간은 입자크기를 증가 시켰다. 코어의 가교는 PMMA 사슬의 움직임을 제한하여 입자크기를 감소시킬 수 있었다. 형광 나노입자는 용매의 증발을 이용한 적재 방법으로 커큐민을 PMMA 코어에 적재하여 제조되었다. 소수성을 가지는 커큐민은 물과 같은 친수성 용매에 녹지 않기 때문에 유기 용매에서와 같은 광학 특성을 나타내기 어려운 단점을 가지고 있다. 커큐민은 양친성 나노입자에 적재됨으로써 단점이 보완되었다. 친수성 PEI 쉘은 커큐민이 적재된 나노입자가 충분히 물에 녹을 수 있게 하여 유기 용매에서와 같은 광학 특성을 나타낼 수 있게 되었다. 나타난 광학 특성은 자외선-가시광선 분광기와 형광 분석기를 통해 확인 하였다. 형광 나노입자는 수용액상에서 가시광선에서 노란색을 띄었고, 자외선 하에서 녹색 형광을 나타냈다. 이것은 유기 용매에서 커큐민의 성질과 같다. 우리는 제안된 방법으로 제조된 형광 고분자 나노입자가 입자크기 조절이 용이하고, 안정하며 수용액상에서 광학적인 특징을 보이는 점에서 화학센서, 전자광학 그리고 생명 공학 등에 영향을 줄 수 있을 것으로 기대한다.

In this study, we prepared fluorescent polymeric core–shell nanoparticles containing biomolecular curcumin. The fluorescent polymeric nanoparticles consist of hydrophilic polyethyleneimine (PEI) as shell materials, hydrophobic poly(methyl methacrylate) (PMMA) as core materials and fluorophore curcumin loaded in the core. The polymeric core–shell nanoparticles were prepared by simple one–pot grafting co–polymerization step of PEI containing the amino group with MMA and single purification step. Prepared nanoparticles have size–controllable character via adjustment of MMA monomer concentration and reaction time. The change of particle size was confirmed by a zeta sizer. The low monomer concentration and short reaction time reduced particle size, but the high monomer concentration and long reaction time increased particle size. The fluorescent core–shell nanoparticles were prepared by loading curcumin in PMMA core through solvent evaporation method. The curcumin which has hydrophobic character has drawback difficult to perform optical properties in the hydrophilic solvent such as water because it cannot be sufficiently dispersed in the water. The drawback of curcumin was redeemed by loading curcumin in amphiphilic PMMA–PEI nanoparticles. The loaded curcumin in PMMA–PEI can perform optical properties because the hydrophilic PEI shell sufficiently makes to soluble in the hydrophilic solvent. The performed optical properties were confirmed by ultraviolet–visible spectroscopy (UV–vis) and photoluminescence spectroscopy (PL). The fluorescent nanoparticles which were dissolved in water showed yellowish color at visible light and greenish fluorescence at UV light. We expect that the fluorescent polymeric nanoparticles prepared by the proposed method can affect chemical sensors, electro–optics, and biotechnology because of easily control the particle size, exhibit stable and optical properties.
