As an alternative, resistive switching random access memory (RRAM) relying upon a switching mechanism based on change in resistance has attracted attentions as a promising next generation nonvolatile memory owing to its simple structure, facile processing, high density, and fast switching speed. To date, various insulating or semiconducting materials have been found to demonstrate resistive switching, including a reduced graphene oxide (rGO) and silk protein. Actually, the rGO can be used as a promising active layer in resistive switching memories as well as flexible electrodes with highly conductive sheets. In addition, silk protein is one of the most widely used bio-compatible materials for bio-implantable device. Recently, it has emerged as a promising candidate for electronic memory devices owing to its excellent physical, chemical and biological properties. On the other hand, for practical application about the resistive switching devices, cortisol detection was pursued in this thesis. The cortisol is a steroid hormone secreted from the adrenal cortex on kidney that plays an important role in increasing blood pressure and glucose levels. The cortisol secretion mainly depends on the level of psychological and emotional stress, and thus it is also known as “stress hormone”. In this thesis, immunosensor using reduced graphene oxide (rGO) and elastin-liked-polypeptide (ELP) as electrical sensing layer based on resistive switching property were demonstrated respectively. To demonstrate immune sensor for cortisol detection based on the resistive switching operation, current/voltage (I-V) characteristics were measured. Specific immune recognition between anti-cortisol monoclonal antibody (c-Mab) and cortisol was examined on a resistance change at the rGO and ELP channel. The sensor was proposed with sensing mechanism based on rGO channel resistive switching device, which was worked with space charged limited current (SCLC) conduction mechanism. Also, the sensor could detect human salivary cortisol levels. For an immune sensor by ELP channel, the bio-engineered ELP consists of protein or peptide synthesized as [Val-Pro-Gly-Val-Gly]32 sequence, which could be used for the memory function. By using ELP active channel, specific immune binding affinity between probe c-Mab and target cortisol was realized by attaining its calibration plot and a limit of detection (LOD) test was performed on a basis of resistance change or resistive switching phenomena by measuring I-V characteristics. The calibration plot of linear region with the cortisol concentration, was used to quantify the concentration of cortisol in real saliva samples. This immuno binding detection method accurately measured cortisol in the collected saliva samples achieved to a concentration of 1 ng/mL (2.76 nM). This study suggests that the resistive switching device would provide a novel application, which will be utilized to point-of-care (POCT) biosensor for in vitro study about stress-related physiology.
