LPS로 처리한 BV-2 세포에서 백하수오 추출물은 NO 생성, iNOSmRNA 및 단백질 생성을 농도 의존적으로 저해하였다. 백하수오 추출물은 이 농도 범위에서 세포 증식에 영향을 주지 않음을 확인하였다. 백하수오 추출물에서 분리한 시난디온 A는 LPS로 처리한 BV-2 세포에서 NO 생성을 강력하게 저해하였으며 IC50 값은 27.1 μM 정도로 확인되었다. 이 물질은 80 μM 농도까지 세포 증식을 거의 저해를 하지 않았다. 시난디온 A는 iNOS mRNA와 단백질 발현뿐 아니라 COX-2 mRNA와 단백질 발현을 저해함을 보여주었다. 이 물질은 또한 염증성 사이토카인인 TNF-α, IL-1β, IL-6 mRNA 발현을 저해함을 보여주었으며, 이들 사이토카인 단백질 생성을 저해함을 ELISA 방법에 의해 보여주었다. 시난디온 A가 항산화 활성이 있는지 라디칼 포식활성측정법에 의해 측정한 결과, 항산화 활성이 우수한 것으로 확인하였다. 시난디온 A는 BV-2 세포 내에서 항산화 활성에 관여하는 heme oxygenase-1 (HO-1) 효소의 mRNA와 단백질 생성을 농도 의존적으로 증가시키는 것을 보여주었다. 세포 내에서 염증 및 산화 반응에 중요한 역할을 하는 NF-κB 전사인사는 LPS에 의해 활성화가 되는데, 시난디온 A는 NF-κB 저해 단백질인 IκB의 LPS에 의해 유도되는 인산화를 저해하여, NF-κB의 핵으로의 이동을 감소시키는 것이 확인되었다. 이 결과는 시난디온 A가 LPS에 의해 활성화되는 NF-κB의 활성을 저해시키고, NF-κB에 의해 전사가 유도되는 iNOS, COX-2, TNF-α 등 사이토카인의 생성을 저해시킨다는 것을 보여준다. 따라서 백하수오의 염증관련 질병에 대한 치료제로의 효능은 많은 기전중의 하나로서 NF-κB 신호전달의 저해에 기인한다는 것을 보여준다.

Cynanchum wilfordii has been used traditionally used in asian countries for centuries for prevention and treatment of numerous diseases. Cynanchum wilfordii extracts (CWE) have been reported to exhibit various biological activities including improvement of menopausal syndrome, improved endothelial dysfunction and protection of neurons from oxidative stress but its anti-inflammatory activities in microglial cells have not been shown yet. In this study, we investigated the anti-inflammatory and antioxidant activities of CWE and cynandione A(CA) isolated from the extract. CWE inhibited LPS-induced NO production and expression of inducible nitric oxide synthase (iNOS) mRNA protein in BV-2 cells. CWE did not affect growth of BV-2 cells within the concentrations used in this study. CA strongly inhibited NO production in LPS-stimulated BV-2 cells and IC50 was 27.1 μM. CA did not show any cytotoxicity up to the concentration of 80 μM. CA was found to reduce LPS-induced expressions of iNOS and cyclooxygenase-2 (COX-2) at the mRNA and protein levels in a dose-dependent manner. CA inhibited LPS-induced mRNA expression of pro-inflammatory cytokines, including TNF-α, IL-1β and IL-6. ELISA analysis showed that CA inhibited the proinflammatory cytokines in LPS-treated BV-2 cells. This study also demonstrated that CA exhibited strong free radical scavenging activity, and induced antioxidant enzyme heme oxygenase-1 (HO-1) gene expression at the mRNA and protein levels in dose-dependent manner. Futhermore, analysis of effects of CA on NF-κB activation showed that CA significantly suppressed the LPS-stimulated phosphorylation of inhibitory kappa B-α (IκB-α) and the subsequent translocation of NF-κB p65 to the nucleus. These results indicated that CA inhibited LPS-induced activation of NF-κB, which in turn suppressed expression of NF-κB-target genes, including iNOS, COX-2, and proinflammatory cytokines such as TNF-α. These studies suggest that Cynanchum wilfordii may be beneficial for treatment of various chronic inflammatory diseases and that the anti-inflammatory activities may be partly due to the inhibition of NF-κB signaling pathway by CA and/or other constituents contained in Cynanchum wilfordii.
