In this study, the antioxidant and anti-allergic effects of Perilla frutescens var. acuta Kudo and its active compound rosmarinic acid were investigated for developing health functional foods. First of all, the extraction method of Perilla frutescens (PF) was optimized, the marker compound, rosmarinic acid in PF extract was determined and its analysis conditions were validated using HPLC and liquid chromatography mass spectrometry(LC/MS). The highest total phenol and flavonoid contents in PF were extracted in a 30% ethanol aqueous solution at 80℃ for 24 hr under reflux extraction methods. The marker compound, rosmarinic acid was stably isolated from PF-30% ethanol extract (PF-E30) at pH 5.5 and content of it in PF-E30 was approximately 12.3 mg/g. Next, the anti-allergic effect of PF-E30 on various allergic reactions using in vitro and in vivo models was examined. PF-E30 significantly reduced the compound 48/80-induced histamine release and anaphylactic reaction. In addition, PF-E30 significantly inhibited the IgE-induced passive cutaneous anaphylaxis in a dose-dependent manner. Finally, the functionality, safety of PF and the intake were examined, and the standards and specification of PF were set. Based on these studies, the author confirmed that the PF has a function of 'effectiveness in improving immune hypersensitivity' as a natural product of functional food. In conclusion, this study shows that the optimal extraction method, reproducible quantitative analysis of PF-E30 and rosmarinic acid (a marker compound of PF-E30) were established. In addition, these findings indicate that PF-E30 has a potent anti-allergic activities as an effective and safe ingredient or functional food in allergic reactions.
