Effect of chinese herbal Paeonia lactiflora & Poria cocos to treat androgenetic alopecial on mice models

Androgenic alopecia is the most common type of alopecia. The symptom of androgenic alopecia are constant hair sparsity and hair follicle atrophy. The parts of the male pattern alopecia are usually the head and frontotemloral, while women are visible hypotrichosis. And both of men and women are all with the hair greasy, dandruff and itching. Currently, many drugs have side effects for the treatment of androgenic alopecia. In order to explore the therapeutic effect of paeonia lactiflora and poria cocos on androgenic alopecia and its related mechanisms, C57BL/6 mice were used as experimental subjects,paeonia lactiflora and poria cocos were given to observe the condition of alopecia. The effects of paeonia lactiflora and poria cocos on androgenic alopecia were studied according to the contents of Triglyceride, total cholesterol, testosterone and estradiol in serum and the ratio of T / E2.
Before the start of the experiment, hair on the back of the mice was removed using a razor and a hair removal cream to induce hair loss models. SHAM group daily 70% enthanol 0.1ml applied to the back skin. Control group daily 0.1ml of 0.5% testosterone propionate solution administered topically on back skin. In the positive control group, 0.1 ml of 2% finasteride solution was applied daily after 0.1 ml of 0.5% testosterone propionate solution was applied for 30 minutes. The paeonia lactiflora group was given daily with a solution of 0.5 ml peony lactiflora extract (1500 mg / kg BW) mixed with 5.5 ml distilled water. Poria cocos group was given daily 0.5ml Poria cocos extract (1500mg / kg BW) and 5.5ml of distilled water to form a solution. Mixed group (Paeonia lactiflora and Poria cocos ratio 1: 1) given 0.5ml paeonia lactiflora and Poria cocos extract, mixed with 5.5ml of distilled water to form a solution. Take pictures and comments every Monday and Thursday.
In weight gain, average dietary intake, food efficiency ratio, there were differences between paeonia lactiflora group and poria cocos and other groups. Among them, in weight gain and average dietary intake, there was significant difference between paeonia lactiflora pall and poria cocos group ( p < 0.05 ), but there was no significant difference between paeonia lactiflora pall and control group and positive-control(p-control) group. There was no significant difference between the group of paeonia lactiflora, poria cocos and control in the food efficiency ratio, but the difference was significant (P<0.05) with the other three groups. In this study, fat was divided into two groups: out fat and in fat. the results showed that there was no significant difference between control group, paeonia lactiflora group and poria cocos group, but there was significant difference with other groups ( p < 0.05 ). In biochemical test, testosterone test results showed that there was a difference between sham group and other groups ( p < 0.05 ), but in the comparison of estradiol, there was no significant difference between the groups. And Moreover, there was no significant difference between the groups in the comparison of The ratio of testosterone to estradiol(T/E2). The test results of Triglycerides showed that there was a difference between the poria cocos group and the other five groups (P<0.05). In the detection of Total cholesterol , there was a difference between peony and poria (P<0.05), and there was no difference between the paeonia lactiflora group and the SHAM group and the P-control group. The results of functional evaluation showed that the survival rate of hair follicle number and hair growth rate in paeonia lactiflora group were similar to those in p - control group and higher than those in other groups ( p < 0.05 ).
Poria cocos can alleviate androgenic alopecia to some extent, but the effect is not obvious. Compared with Poria cocos, paeonia lactiflora has a more significant effect. Compared with the finasteride used in the positive control group, the effect was comparable. But the paeonia lactiflora does not alleviate and treat alopecia by improving the testosterone content of the experimental animals. The specific mechanism of treatment needs to be further explored.
