Cells starved with glucose may require high mitochondrial ATP production and activated autophagy. Here, I examined changes in status of mitochondria and autophagy in cells that have two contradictory demands, i.e., increase in oxidative phosphorylation and autophagic supply of glucose. Under glucose starvation, total cellular ATP level was increased. In addition, mitochondria ATP production was accelerated, which is mainly attributed to increased mitochondria content. Mitochondrial biogenesis upregulation and elongated structure, inadequate structure for mitophagy, were seemed to be responsible for increase of mitochondria content. Together with preservation of mitochondria for respiration, autophagy initiation was indeed activated while autophagy flux was blocked. Autophagic flux can be impaired by problem of fusion of autophagosome and lysosome, but autophagosome was fused with lysosome notably. However, I observed abnormally large autophagosomes while formation of autolysosome was executed normally. Number of lysosome was increased, but not all active, and lysosomal acidity was diminished even though lysosome biogenesis was increased. These observations indicate that abnormal autophagosomes were accumulated as autolysosomes with improper degradation. Nevertheless, I found that mitophagy could be executed even when autophagic flux was blocked in glucose-starved cells. The results of my study suggest that, under glucose starvation, mitochondria were preserved through increased biogenesis and elongated structure, and this result in increase of mitochondrial ATP production, leading to increase of total ATP level. In addition, autophagy was activated but autophagic flux was impaired by defect in lysosomal degradation.
