Together with production of functional mitochondria, structural dynamics of mitochondria and removal of the damaged mitochondria through mitophagy are believed to be critical for maintaining mitochondrial integrity. We assumed that glucose starvation is possible way to enhance the mitochondrial turnover through metabolic means. It is also expected to shift cellular reliance on glycolysis to mitochondrial respiration for ATP production, and therefore, to bring in a substantial alteration in the function and status of mitochondria. When fibroblasts were cultured in a medium devoid of glucose, the content dropped down rapidly. But, it was increased gradually after 3 hr and almost completely restored at 12 hr and further increased thereafter. The mRNA levels of mitochondrial proteins and mitochondrial biogenesis proteins dropped down with a kinetics similar to that of the mitochondrial content indicating that this decrease is largely due to the low mitochondria biogenesis. After 10 hr, mitochondria became filamentous forming networks despite autophagic activation. We speculate that mitophagy is attenuated due to the formation of elongated mitochondria. Interestingly, cellular ATP level did not decrease during glucose starvation. Overall, in glucose starvation, cells maintain mitochondria at high level in order to keep efficient ATP production by altering the mitochondria structural dynamics even though they need to activate autophagy.
