DA-9805 is a new botanical antiparkinsonism drug candidate formulated with ethanolic extract of root of Bupleurum falcatum, root cortex of Paeonia suffruticosa, and root of Angelica dahurica. To determine saikosaponin a (SSa), paeonol (PA), and imperatorin (IMP), which are the three active/representative ingredients of DA-9805, in rat plasma simultaneously, a sensitive and rapid LC-MS/MS method has been developed. A calibration curve was constructed in the range of 0.5‒1000 ng/mL for SSa, 20‒10000 ng/mL for PA, and 0.2‒1000 ng/mL for IMP. All the validation data, including the selectivity, linearity, precision, accuracy, recovery, matrix effect, and stability satisfied the acceptance requirements. This method was applied in pharmacokinetic studies of SSa (1.1‒4.6 mg/kg), PA (14.8‒59.2 mg/kg), and IMP (1.4‒11.5 mg/kg) following intravenous and oral administration of each pure component at various doses or equivalent doses of DA-9805. Following intravenous administration of each component, all the three components showed greater dose-normalized AUC and slower CL following intravenous administration of the higher dose. In contrast, dose-proportional AUC values of SSa, PA, and IMP were observed following oral administration of each pure components or DA-9805. Compared to oral administration of each pure compound, oral administration as DA-9805 showed increase in AUC of SSa (by 96.1‒163%) and PA (by 155‒164%) possibly due to inhibition of their metabolism by IMP. Delay in absorption of PA and IMP following DA-9805 administration was also observed compared with pure compound administration. Binding values of SSa and IMP to plasma protein or brain homogenates were considerable, whereas PA have relatively low binding values. All the three components of DA-9805 showed greater binding values in the brain homogenates compared to those in plasma, which might enable the brain to plasma ratios greater than unity following multiple oral administration of DA-9805 (250 mg/kg/day for 8 days). In contrast, negligible levels of them in cerebrospinal fluid (CSF) were observed following the multiple oral administration of DA-9805. Drug interaction potential of DA-9805 (single oral dose of 60 mg/kg or multiple oral doses of 60 mg/kg/day for 7 days) evaluated with oral cocktail probe substrates of cytochrome P450 (CYP) 1A2, 2B6, and 3A subfamily suggested no significant effects of oral DA-9805 on activities of CYP1A2 and 2B6, whereas slight inhibitory effect on CYP3A activity was observed. However, this inhibitory effect of DA-9805 on CYP3A activity may not be considerable in effective dose ranges of DA-9805. This dissertation contributes to better understanding of the comprehensive pharmacokinetic characteristics of SSa, PA, and IMP in rats, comparative oral pharmacokinetics between each pure component and DA-9805, and in vivo drug interaction potential of DA-9805 as a predator.
