Bladder cancer is one of the tumors associated with the highest mortality rate. It is clinically characterized by high recurrent rates and poor prognosis once tumors invade the lamina propia. Nevertheless, current diagnostic techniques such as cystocopy and cytology are invasive, time-consuming, and expensive. To address these challenges, we developed an electrochemical enzyme-linked immunosorbent assay (eELISA) platform on the surface of indium tin oxide (ITO) to quantify matrix metalloproteinase-9 (MMP-9) proteins and apolipoprotein-A4 (Apo-A4) proteins, reported as bladder cancer biomarker in urine with high sensitivity and specificity. Combined with cyclic voltammetry (CV) method, we developed ITO-based eELISA platform and it successfully determined the existence of MMP-9 and Apo-A4 as low as 0.33 ng/mL and 11.4 ng/mL with the dynamic range of 0.4- 100 ng/mL and 25- 250 ng/mL, respectively. Thus, the method provided the calibration curve of MMP-9 and Apo-A4 under the electrochemical approach. We expect that our developed eELISA platform would offer a simple and sensitive immunoassay approach for point-of-care (POC) diagnostic system.
