근육량 또는 근기능의 감소를 특징으로 하는 근감소증은 근육의 미사용, 노화, 그리고 만성질환에 의해 일어난다. 특히, 사코페니아(sarcopenia)라고 불리는 노화성 근감소증은 노인들의 근육 기능을 떨어뜨리고 미토콘드리아에 악영향을 준다. 국화(Chrysanthemum morifolium) 추출물(CME)과 이의 활성 성분인 isochlorogenic acid A (IcA)는 항산화, 항염증, 항당뇨 및 신경보호 효과가 있는 것으로 보고 되어 있다. 그러나 현재까지 노화성 근감소증에 대한 억제 효능은 밝혀진 바가 없다. 이에 따라 본 연구에서는 CME와 IcA가 단백질 전환과 미토콘드리아 생합성을 조절함으로써 근육의 양과 기능의 감소를 억제할 수 있는지에 대하여 연구를 진행하였다. 동물 모델에서, CME와 IcA는 근육의 질량, 부피 및 단면적의 크기를 증가시켰고, 게다가, 근력과 유산소 운동 능력을 향상 시켰다. 분자기전의 측면에서 보았을 때, CME와 IcA는 단백질 분해와 관련이 있는 muscle atrophy F-box (MAFbx also called atrogin-1)와 muscle ring-finger protein 1 (MuRF-1)의 발현을 억제하였을 뿐만 아니라, 단백질 합성과 연관이 있는 phosphatidylinositol 3 kinase (PI3K)/Akt 신호전달체계를 활성화함으로써, mammalian target of rapamycin (mTOR)/70-kDa ribosomal protein kinase S6 (p70S6K)/eukaryotic initiation factor 4-binding protein 1 (4EBP-1) 신호전달체계의 활성화에 영향을 미쳤다. 또한, CME와 IcA는 미토콘드리아 생합성과 관련된 인자인 peroxisome proliferator-activated receptor-gamma coactivator-1 alpha (PGC-1α), estrogen-related receptor alpha (ERRα), mitochondrial transcription factor A (Tfam) 및 nuclear respiratory factor-1 (NRF-1)의 발현을 증가시켰다. 특히, CME와 IcA는 미토콘드리아 양을 증가시키고, 동시에 미토콘드리아에 있는 시트르산합성효소 활성과 adenosine triphosphate (ATP) 생성을 증가시켜 기능성을 향상시켰다. 이러한 결과들은 CME와 IcA가 노화에 의한 근육량 및 근기능 감소에 대한 억제 효과를 갖는 기능성 식품 또는 약품 소재로써 사용될 수 있음을 시사한다.

Especially, age-related muscle atrophy, called sarcopenia, results in deterioration of muscle function and mitochondria in older people. Chrysanthemum morifolium Ramat. extract (CME) and isochlorogenic acid A (IcA) have been reported to have anti-oxidant, anti-inflammatory, anti-diabetes, and neuroprotective activities. However, inhibitory effects of CME and IcA on sarcopenia have not been studied yet. This study evaluated whether CME and IcA delayed loss of muscle mass and function, by regulating protein turnover and mitochondrial biogenesis in L6 skeletal muscle cells and middle-aged mice. In animal experiment, CME and IcA increased muscle mass, volume, and fiber. In addition, CME and IcA significantly enhanced grip strength and exercise endurance. At the molecular level in both of in vitro and in vivo, CME and IcA not only prevented muscle atrophy F-box (MAFbx, also called atrogin-1) and muscle RING-finger protein-1 (MuRF-1) associated with proteolysis but also affected mammalian target of rapamycin (mTOR)/70-kDa ribosomal protein kinase S6 (p70S6K)/eukaryotic initiation factor 4-binding protein 1 (4EBP-1) pathway for protein synthesis by activating phosphatidylinositol 3-kinase (PI3K)/Akt pathway. Moreover, CME and IcA up-regulated mitochondrial biogenesis-related factors, including peroxisome proliferator-activated receptor-gamma coactivator-1 alpha (PGC-1α), estrogen-related receptor alpha (ERRα), mitochondrial transcription factor A (Tfam), and nuclear respiratory factor-1 (NRF-1). Particularly, CME and IcA increased mitochondrial density and functions, such as citrate synthase activity and the adenosine triphosphate (ATP) production in mitochondria matrix. These results suggest that CME and IcA can be resources of functional food or medicine having inhibitory effect on sarcopenia.
