Background and Purpose
Celastrol had the greatest enhancing effect on ionizing radiation-induced cell death, and show potentiate apoptosis in numerous tumor cells.
1H magnetic resonance pectroscopy(MRS) has been suggested as a measure of therapy-induced changes at the cellular level such as necrosis, apoptosis and cytotoxic edema because viable cells act as a diffusion barrier in tissue.
In this study was to assess the potential of tumor response for combining Celastrol with radiotherapy (RT) using noninvasive MRI techniques.

Materials and Methods
Three groups (n=6 per group) of A549 human lung carcinoma murine model were consisted of RT (10 Gy) group, Celastrol (2 mg/kg/d) group and combined Celastrol with RT group. Metabolic change was evaluated by using MRS was obtained by using point resolved spectroscopy (PRESS) sequence. MRS were performed before and on the day 3, and 6 after therapy initiation. All tumors were collected on the day 6 post therapy for hematoxylin and eosin (H&E) staining. The MR spectrum was assessed with repeated measure ANOVA and Tukey’s post-hoc test. The H&E staining analysis for the area of necrosis was assessed with one-way ANOVA and post-hoc Tukey’s test.

Results
Metabolic change of combined Celastrol with RT group significantly decreased at the day 3 and day 6 (p<0.05). Metabolic change of celastrol only group is not significantly decreased on the day 3 and day 6. RT only group is not significantly decreased on the day 3 and day 6.


Conclusion
Our results suggested that combined therapy of Celastrol with RT was more effective antitumor therapy than that of RT or Celastrol therapy. 1H MR Spectroscopy would be a potential tool for assessing tumor response for combining Celastrol with RT in human lung cancer model.
