Peroxisomes are dynamic organelles that participate in a diverse array of cellular processes, including β-oxidation of branched-chain and very-long-chain fatty acids, which results in reactive oxygen species (ROS) production. To counterpoise the damaging effect of ROS, the antioxidant peroxisomal enzyme catalase reduces H2O2 to water and oxygen. The present study investigated the role of catalase in peroxisome degradation during nutrient deprivation. Short interfering RNA-mediated silencing of catalase and its pharmacological inhibition by 3-aminotriazole (3AT) decreased the number of peroxisomes and resulted in the downregulation of peroxisomal proteins The present study also investigated the role of catalase in liver function and lipid metabolism during nutrient deprivation using catalase knockout (KO) mice. We found that catalase KO in mice caused elevated levels of serum aspartate transaminase and alanine transaminase during starvation, indicating that catalase plays a critical role in preventing liver damage during starvation. Consistent with ROS decay being mediated by catalase, catalase depletion during starvation triggered dramatic increases in ROS and inflammation, which may result in increased apoptotic cell death. Catalase depletion during starvation also decreased peroxisomal protein, which resulted in pexophagy. Unexpectedly, we found that hepatic and serum levels of triglycerides were dramatically decreased in catalase KO mice during starvation, which is potentially associated with the elevated fat consumption through the activation of lipolysis and UCP1 in adipocytes during fasting. These findings suggest that catalase plays an important role in pexophagy in the liver and HepG2 cell during nutrient deprivation. These finding also demonstrate the role of catalase in liver function and lipid metabolism during nutrient deprivation.
