In this study, physical properties and drug release patterns of hydrogel contact lenses prepared as drug delivery media were examined. The tear pH of normal subjects is similar to that of blood, and the physiological pH of tears varies between pH 5.8 and 8.35 due to environmental and physical influences. The drug release behavior of drug delivery contact lenses was analyzed with reference to the pH environmental conditions of these tears. In the preparation of hydrogel contact lenses, hydrogel contact lenses were prepared by different polymerization ratios of 2-hydroxyethyl methacrylate (HEMA) and anionic functional groups (VP, vinylpyrrolidone) and cationic functional groups (NIPAAm, N-isopropylacrylamide). The drug delivery contact lens was prepared by immersing a hydrogel contact lens made in HPMC (hydroxypropyl methylcellulose) drug for 3 minutes. The drug (HPMC) is known to be excellent in the treatment and prevention of cell proliferation, corneal protection, and dry eye syndrome on the corneal surface, because of the constant drug delivery rate regardless of pH change, this drug was used in the study of the drug delivery lens. The surface and pore size of the lens were observed using an optical microscope and a scanning electron microscope (SEM). The particle size analyzer measured the particle size of the dissolved drug (HPMC). The transmittance of UV-B, UV-A, and Visible was measured using a light transmittance meter (TM-1). The outward diffused drug release was analyzed by measuring the optical density using a UV-Spectrophotometer and converting it into the amount of drug. To investigate the reproducibility of pH - responsive hydrogel in drug delivery contact lenses, acidic solution (pH 5.80) and basic solution (pH 8.03) were prepared and analyzed for drug release tendency. As a result, the lens structure of poly(HEMA-co-VP) lens was expanded as the pH increased. Also, it was confirmed that as the polymerization ratio of the -VP functional group increases, the pore of the lens increases. The 20 wt% poly(HEMA-co-VP) lens showed the water content of 17.71% higher than the 0 wt% poly(HEMA-co-VP) lens. In the poly(HEMA-co-NIPAAm) lens, as the pH increased, the lens shrank and the water content decreased. The cationic lens (poly(HEMA-co-NIPAAm)) means that the pore is reduced when the external pH is increased, which means that the poly(HEMA-co-NIPAAm) lens exhibits a higher water content at weak acidity (pH 5.80). The poly(HEMA-co-NIPAAm) 20 wt% lens showed 9.93% lower water content in weak basicity than weak acidity. In the basic (pH 8.03), the poly(HEMA-co-VP) (20 wt%) lens showed a 3.50-fold increase in drug release compared to the mild acidity (pH 5.80), but the poly(HEMA-co-NIPAAm) and the amount of drug released was smaller than that of a weakly acidic lens. The anionic lens, poly(HEMA) 100 wt% lens and poly(HEMA-co-VP) lens, were found to release more drug as the solution pH increased. Conversely, the poly(HEMA-co-NIPAAm) lens, a cationic lens, was found to release more drug in weakly acidic solution (pH 5.80) than weakly basic solution (pH 8.03). In addition, when the poly(HEMA) 100 wt% lens and the poly(HEMA-co-VP) lens were compared, the same anionic lens showed different drug release patterns. For the poly(HEMA) 100 wt% lenses, rapid release rate changes were observed between pH 5.80 and pH 6.45. On the other hand, in the poly(HEMA-co-VP) lens, the release rate of the drug rapidly increased at pH 7.20 and pH 8.03. At the pH of all solutions, lenses with ionic functional groups (-VP, -NIPAAm) exhibited more cumulative emissions than poly(HEMA) 100 wt% lenses. In particular, the poly(HEMA-co-NIPAAm) 5 wt% lens showed a cumulative emission of 5.93 times more than the poly(HEMA) 100 wt% lens, despite its low compounding concentration. All types of lenses were found to be consistent in drug release pattern in alternating immersion of acid solution (pH 5.80) and basic solution (pH 8.03). The drug release was activated at pH 8.03 for anionic (poly(HEMA) 100 wt%, poly(HEMA-co-VP) 10 wt%) lenses and at pH 5.80 for cationic (poly(HEMA-co-NIPAAm) 10 wt% Release was activated. In addition, the amount of drug released from acidic and basic solutions in all three lenses was not significantly different. It was found that the immersion period was constant at 30 minutes, and the change of the release rate was maintained with the periodic alternation of pH. The prepared drug delivery contact lenses exhibited different water content, drug release and drug diffusion coefficients, which were the result of the ionization reaction between the ionic groups in the lens and the pH of the solution. These interactions suggest the possibility of designing selective drug release according to the physiological pH of the tears in the production of contact lenses for drug delivery.
