비만은 만성질환 중의 하나로, 전 세계 적으로 기하급수 적인 속도로 증가하고 있으며, 현대 사회에서 주요한 관심사가 되고 있다. 우리 몸에서 음식물 섭취와 에너지 소비의 균형 시스템이 깨지면 비만이 발생하게 되는데 이때, 비만은 제 2형 당뇨병, 고혈당증, 및 죽상 동맥 경화증과 같은 대사성 질환과 연관되어 있다. 예로부터, 해조류에 존재하는 파이토케미컬들이 항비만, 항당뇨, 항염증 등과 관련된 효과가 알려져 있다. 본 연구에서 우리는 해조류 추출물 중 하나인 우뭇가사리 추출물 (GEE)을 이용하여 in vitro, in vivo 모델에서의 지방 축적과 에너지 대사 및 제 2형 당뇨병에 갖는 효과에 대해 평가하였다. 그 결과, GEE는 지방세포에서 C / EBP α와 PPAR γ 를 포함하는 지방 생성 인자를 통해 adipogenesis를 억제함은 물론, 3T3-L1 세포와 고지방식에 의한 비만 유도 마우스에서 UCP-1 및 PRDM16을 통해 백색지방의 갈색 지방화를 통해 에너지 대사를 높여줄 뿐만 아니라 갈색지방의 에너지 대사 활성을 통해 지방 축적이 또한 억제됨을 확인 하였다. 마지막으로, GEE는 당뇨 모델 마우스에서 GLUT4, IRS-1, PI3K, 및 Akt의 활성을 통해 혈액 속의 당을 세포로 흡수를 촉진시킴은 물론 MAPK의 활성을 억제하여 당뇨 합병증을 완화시킴을 확인하였다. 위의 연구결과, GEE가 갖는 지방 축적 억제 및 당 흡수 효과는 비만, 당뇨, 및 당뇨 합병증을 개선하는데 있어 유용하게 활용될 수 있을 것으로 사료된다.

The incidence of obesity is rising at an alarming rate throughout the world and is becoming a major public health concern. Our body then initiates appropriate alterations in food intake and energy expenditure with the ultimate goal of maintaining energy balance. Obesity develops when this system malfunctions. Because obesity is linked metabolic disease such as a type 2 diabetes, hyperglycemia, and atherosclerosis. Numerous studies have indicated that seaweeds and their derivatives may also have potential therapeutic implications for obesity and diabetes. In this study, we evaluate the effect of Gelidium elegans extract (GEE) on anti-obesity and anti-diabetic effect in vitro and in vivo. GEE regulates adipogenesis through the adipogenic factors including C/EBP α and PPAR γ in 3T3-L1 cells. Furthermore, GEE alters browning in white adipocytes as well as regulates adipogenesis-related factors through UCP-1, and PRDM16 in 3T3-L1 cell and high fat diet induced mice. We also found that administered 200 mg/kg/day GEE significantly activated the insulin signaling pathway, as indicated by the levels of GLUT4, IRS-1, PI3K, and Akt compared to corresponding control group. Regarding MAPK signaling pathway such as JNK, ERK1/2, and p38 MAPK, were effectively reduced which is related with diabetic symptoms in GEE-treated groups. Our results suggest that GEE promising functional food applications for prevention of obesity, diabetes, and diabetic symptoms.
