Seapolynol (SN) is a polyphenol mixture derived from Ecklonia cava, and dieckol (DK) is a main compound of SN. SN and DK effectively inhibited lipid accumulation in three experimental models by suppressing adipogenic factors. This SN and DK-induced inhibition of adipogenic factors was shown to be due to the regulatory effect of SN and DK on early adipogenic factors; SN and DK downregulated the expression of Krueppel-like factor 4 (KLF4), KLF5, CCAAT-enhancer-binding protein β (C/EBPβ), C/EBPδ, and Protein C-ets-2 (ETS2), while KLF2, an anti-early adipogenic factor, were upregulated by SN and DK. These results were reflected in down-regulation of late adipogenic factors, such as peroxisome proliferator-activated receptorγ (PPARγ) and C/EBPα, resulting in a decrease in triacylglycerol content. Also, SN and DK-mediated inhibition in early adipogenesis were closely correlated with the inhibition of mitotic clonal expansion via cell cycle arrest. In a zebrafish study, SN and DK were downregulated mRNA expression of C/EBPa, PPARg, fabp11a and srebf1 in zebrafish. In a mouse study, SN and DK effectively reduced body weight and plasma lipid increases induced by a high-fat diet; triglycerides, total cholesterol, and low-density lipoprotein (LDL) levels were markedly reduced by SN and DK. Moreover, SN and DK activated AMP-activated protein kinase-α (AMPKα), an energy sensor, to suppress acetyl-coA carboxylase (ACC), inhibiting lipid synthesis. We also used C57BL/KsJ-db/db mice for evaluation of antidiabetic effect by SN and DK. SN and DK supplementation significantly reduced fasting blood glucose and plasma insulin levels. Furthermore, the administration of SN and DK increaed IRS-1/PI3K/AKT signaling pathway for improvement of insulin sensitivity in db/db mice. In this study suggests that SN and DK may be an edible agent that can play a positive role in prevention of obsity and type 2 diabetes.
