Endl., Thuja orientalis L., Juniperus chinensis ‘Kaizuka’)과 유칼립투스 4종(Eucalyptus cinerea, E. gunnii, E. polyanthemos, E. pulverulenta‘Babyblue’)을 대상으로 방향화합물 중 안정 효과가 높다고 알려진 monoterpene류 6종(α-pinene, β-pinene, 1,4-cineole, 1,8-cineole, limonene, linalool)의 함량을 분석하였다. 그 결과 기능성 방향화합물의 전체함량은 화합물별 차이는 있었으나 침엽수에서는 전나무가 가장 많았으며(α-pinene, 1,4-cineole, limonene), 이어 소나무, 편백 순이었다. 향나무는 리나룰(linalool) 성분이 가장 많았고, 유칼립투스는 E. pulverulenta ‘Babyblue’에서 α-pinene과 1,8-cineole 함량이 가장 많았다.
2. 기능성 방향화합물의 스트레스완화와 혈관기능개선효과 향기성분분석을 통해 α-pinene, β-pinene, limonene 등을 다량 함유한 것으로 알려진 전나무(Abies holophylla Maxim.)의 향이 건강한 30-40대 직장인의 스트레스완화와 혈관기능개선 및 자율신경계에 미치는 영향을 알아보았다. 피험자들은 실험 코디네이터에 의해 각 실험(식물향기흡입 및 무취공기흡입)에 무작위 순서로 배치되었으며, 무작위 배치, 교차설계(randomized, cross-over design)를 이용하여 단일맹검법(single-blind test)으로 진행되었다. 종속변인의 시간별 변화는 향기흡입 시 무취흡입 시보다 수축기혈압(SBP, 7.9%,p=.001), 이완기혈압(DBP, 4.4%, p=.004), 중심동맥혈압(cSBP, 7.3%, p=.001), cDBP, 4.1%, p=.015), 심박수(HR, 5.4%, p<.001) 모두 유의하게 감소하였으며, 동맥경직도를 나타내는 AIx(56.1%, p<.001)와 AIx@75 (103.8%, p=.001) 역시 향기흡입 시 더욱 감소하였다. 이완기혈압을 제외하고 모든 종속변인에서 처리간 유의한 상호작용효과를 보였다. 심박변이도(HRV, 51.6%, p=.006)와 혈관이완능(FMD, 14.03%, p=.039) 역시 무취흡입 시 보다 향기흡입 시 자율신경계와 혈관기능에 긍정적 영향이 확인되었다. 스트레스반응과 회복속도에서 전나무 잎의 향기흡입은 산술테스트 시 스트레스반응을 완화시켰으며 회복속도에도 긍정적 영향을 미쳤다. 스트레스유발 뒤 중심동맥 이완기혈압(cDBP; Cont 14.4%, Treat 14.7%)을 제외하고, 수축기혈압(SBP; Cont 14.3%, Treat 5.8%)과 이완기혈압(DBP; Cont 14.4%, Treat 9.4%), 중심동맥 수축기혈압(cSBP; Cont 14.7%, Treat 9.2%), 심박수(HR; Cont 14.6%, Treat 11.7%)에서 모두 무취공기흡입(Cont) 시 보다 향기흡입(Treat) 시 덜 증가하였으며, 모두 유의성이 인정되었다(p<.001). 자율신경계 지표인 심박수변이도 HRV(heart rate variability)는 무취흡입 시 LF/HF ratio가 45%증가하였으나, 향기흡입 시 LF/HF ratio는 6.8% 감소(p=.011)하여 무취공기흡입 시 보다 스트레스 반응이 완화되었으며, 혈관기능의 변화 역시 향기흡입 시 AIx(Cont 34.7%, Treat 6.6%, p=.053)와 AIx@75(Cont 143.7%, Treat 89.9%, p=.008) 모두 무취흡입 시 보다 덜 증가하였다. 이들 모든 종속변인에서 회복속도 역시 cDBP를 제외하고 향기흡입 시 회복속도가 더 빨랐다. 이와 같은 결과는 전나무 잎의 향이 스트레스 반응을 완화시키며, 빠른 회복을 통해 안정화하는 회복력과 스트레스 저항력의 상승에 영향을 주었음을 의미한다. 이를 통해, 식물의 향을 이용한 원예치료의 적용이 자율신경계 안정을 통해 심리적 안정감과 편안함을 주고, 혈관기능을 개선해 고혈압 또는 심혈관관련 질환 환자의 증상완화에 도움을 줄 것이며, 기능성 방향화합물을 함유한 식물의 선택적 사용이 보다 전문적 원예치료의 적용에 있어 필요함을 확인하였다.
3. 전나무와 유칼립투스에서 기능성 방향화합물의 생성량 증가 Eucalyptus pulverulenta‘Babyblue’와 Abies holophylla Maxim. 잎에서 스트레스완화, 진정효과 및 혈관기능개선 효과가 큰α-pinene과 1,8-cineole, limonene 등의 monoterpene 함량변화에 미치는H2O2와 Salicylic acid (SA)처리의 영향에 대해 알아보았다. 유칼립투스 가지를 H2O2 수용액(0.3, 0.5, 1%)과 SA 수용액(0.1, 1 mM)에 침지하거나 잎에 엽면살포(0.1 mM SA, 1% H2O2)하고, 전나무가지를 1% H2O2 수용액과 0.1 mM SA수용액에 침지처리한 후 시간흐름(0, 0.5, 1, 2, 4시간)에 따른 α-pinene과 1,8-cineole, limonene 등의 monoterpene 함량변화를 분석하였다. 침지처리에서 유칼립투스 잎의 α-pinene 함량(mg.L-1)은 0.1 mM SA에 2시간 침지 후 1.62에서 5.48로 크게 증가하였으며(238.27%, p=0.012), 1,8-cineole 함량(mg.L-1)은 1 mM SA에 4시간 침지 후 44.44에서 78.96으로 가장 크게 증가하였다(77.66%, p=.026). 살포처리에서는 0.1 mM SA수용액을 유칼립투스 잎에 엽면살포 30분 후α-pinene 함량(mg.L-1)은 1.62에서 3.91(141%, p=0.007)과 1,8-cineole 함량(mg.L-1)은 44.44에서 87.91로 증가하였다(98%, p=.010). α-pinene과 1,8-cineole 모두 살포 30분 뒤 크게 증가하여, 엽면살포가 짧은 시간 내에 방향화합물을 증가시키는데 침지처리보다 효과적이었다. 전나무 잎은 α-pinene과 limonene 모두 1% H2O2 수용액에 침지처리 시 30분부터 점차적으로 증가하여 4시간 뒤 각각 2.72±0.27에서 8.15±0.31로(200%, p<.001)와 1.00±0.24에서 6.53±0.45(553%, p<.001)로 크게 증가하였다. H2O2와 SA 처리는 α-pinene과 1,8-cineole, limonene 등의 monoterpene 함량증가에 유의한 영향이 있으며, 유칼립투스에서는 SA의 처리가, 전나무에서는 H2O2처리가 방향화합물 증가에 더 효과적이었다. 전나무 가지를 원예치료 프로그램 4시간 전에 1% H2O2 수용액에 침지하거나, 유칼립투스 가지를 원예치료 2시간 전 0.1 mM SA 수용액에 침지하고, 30분전에 잎에 살포하면 스트레스 감소 및 진정에 도움이 되는 monoterpene의 식물체내 함량이 크게 증가되었다.

Plant fragrance is composed of highly volatile aromatic compounds, among which certain fragrance compounds have therapeutic effects. If these therapeutic effects are used by function, it will be helpful to increase the effect of horticultural therapy. Therefore, it is necessary to analyze the fragrance compounds of plants and to objectively verify the therapeutic effect of using them. In this study, we focused on the fragrance of these plants and investigated the content of functional aromatic compounds in plants with therapeutic effect and the way to increase them. In order to enhance the therapeutic effect of horticultural therapy using plants, the therapeutic effects of fragrance compounds were verified by stress relief, improvement of vascular function and changes of autonomic nervous system.
1. Analysis of Fragrance Components and Functional Fragrance Components of Plants In the horticultural therapy activity, the effect of the mental and physical stability and the stress reduction through the fragrance of plants, which is one of the functional elements of the plant, is expected to play a major role as another added value of plant. This study was conducted to select plants with high content 6 monoterpenes (α-pinene, β-pinene, 1,4-cineole, 1,8-cineole, limonene, linalool) that are known to have high stabilizing effects and apply them to horticultural therapy. For this purpose, monoterpene contents from five species of conifer (two species of Pinaceae, three species of Cupressaceae) and 4 Eucalyptus species (Eucalyptus cinerea, E. gunnii, E. polyanthemos and E. pulverulenta‘Babyblue’) which are widely used as green materials in flower design work and horticultural therapy activities were analyzed. As a result, the content of monoterpenes with therapeutic effect was the highest in the fir trees, and followed by pine trees and cypress trees with the difference between components. The content of linalool was highest in juniper trees. In Eucalyptus, Eucalyptus pulverulenta‘Babyblue’ contained the highest contents of α-pinene and 1,8-cineole.
2. Effects of Fragrance Components of Abies holophylla Max. Stress Relief, Vascular Function and Autonomic Nervous System The purpose of this study was to investigate the effects of fragrance of Abies holophylla Max., which is known to contain α-pinene, β-pinene, limonene, etc. on stress relief, vascular function and autonomic nervous system. In this cross-over design study, 15 healthy workers without otolaryngologic disease (37.6 ± 6.8 years) were randomized to either (1) plant fragrance inhalation or (2) odorless air inhalation by an experiment coordinator. Compared with odorless air inhalation, plant fragrance inhalation decreased brachial SBP from 117.64±4.89 to 108.36±3.92 mmHg (7.9%, p=.001), brachial DBP from 75.00±3.21 to 71.71±2.30 mmHg (4.4%, p=.004), central SBP from 104.86±4.22 to 97.21±3.28 mmHg (7.3%, p=.001), central DBP from 75.00±3.14 to 71.93±2.43 mmHg (4.1%, p=.015), and heart rate from 66.57±1.84 to 63.00±1.72 bpm (5.4%, p<.001). Arterial Stiffness Indexes such as AIx from 13.00±3.75 to 5.71±4.19% (56.1%, p<.001) and AIx@75 (7.71±3.51 to –0.29±3.93% (103.8%, p=.001) were also decreased significantly when fragrance was inhaled. There was a significant interaction effect between all treatments except diastolic blood pressure. Heart rate variability (HRV, 51.6%, p=.006) and flow-mediated dilation (FMD, 14.03%, p=.039) were significantly improved in plant fragrance inhalation vs. odorless air inhalation. In the stress response and recovery rate, the fragrance inhalation of the fir leaf alleviated the stress response in the arithmetic test and increased recovery rate. After stress, brachial SBP(Cont 14.3%, Treat 5.8%), brachial DBP(Cont 14.4%, Treat 9.4%), central SBP(Cont 14.7%, Treat 9.2%), and HR(Cont 14.6%, Treat 11.7%) were lower in fragrance inhalation(Treat) than those of odorless air inhalation(Cont) (p<.001), except for the central DBP(Cont 14.4%, Treat 14.7%). HRV (heart rate variability), which is an indicator of autonomic nervous system, was lowered in the LF/HF ratio (6.8%, p=.011) at the time of fragrance inhalation and the stress response was alleviated at the time of inhaling odorless air. Changes in vascular function were also less in AIx (Cont 34.7%, Treat 6.6%, p=.053) and AIx@75 (Cont 143.7%, Treat 89.9%, p=.008) than in the odorless inhalation. In addition, recovery rates were faster in all dependent variables except for cDBP.In conclusion, this study demonstrate that fragrance components of Abies holophylla Max. have positive effects of autonomic nervous system and improve vascular function. Therefore, it will be helpful for the selective use of plants to increase the treatment effect in horticultural therapy.
3. Effects of H2O2 and Salicylic Acid Treatment on the Monoterpene Content of Eucalyptus pulverulenta‘Babyblue’and Abies holophylla Maxim. This study was conducted to investigate the effect of hydrogen peroxide (H2O2) and salicylic acid (SA), treatment on the increase of monoterpene contents such as α-pinene, limonene and 1,8-cineole in Eucalyptus pulverulenta‘Babyblue’ and Abies holophylla Maxim. E. pulverulenta‘Babyblue’ shoots were soaked in various concentrations of chemical solution (0.3%, 0.5%, 1% H2O2 or 0.1 mM, 1 mM SA) or its leaves were sprayed with a 1% H2O2 or 0.1 mM SA solution. These were then analyzed for there α-pinene and 1,8-cineole contents at various periods after treatment (0, 0.5, 1, 2, and 4 h). A. holophylla Maxim. was soaked in 1% H2O2 or 0.1 mM SA solution then analyzed for the contents of α-pinene and limonene for 0, 0.5, 1, 2, and 4 hours after treatment, respectively. After 2 h of soaking, the α-pinene content in the 0.1 mM SA solution increased from 1.62 to 5.48 mg.L-1 (238%, p=.012), whereas the 1,8-cineole content in the 1 mM SA solution increased from 44.44 to 78.96 mg.L-1 (78%, p=.026) after 4 h of soaking. In response to spraying 0.1 mM SA on leaves, after 30 min, the α-pinene content increased from 1.62 to 3.91 mg.L-1 (141%, p=.007), whereas the 1,8-cineole content increased from 44.44 to 87.91 mg.L-1 (98%, p=.001), which means both increases were significant. From soaking treatment in A. holophylla Maxim, the content of α-pinene increased from 2.72 to 8.15 mg.L-1 (200%, p<.001) with soaking for 4 hours in 1% H2O2 and the limonene content increased from 1.00 to 6.53 mg.L-1 (553%, p<.001) with soaking for 4 hours in 1% H2O2. From these results, it was recommended that the soaking of A. holophylla Maxim. in 1% H2O2 solution 4 hours and E. pulverulenta‘Babyblue’ shoot in 0.1 mM SA solution 2 hours, or spraying with 0.1 mM SA solution 30 minutes before horticultural therapy to increase the content of monoterpenes which reduce stress, soothes and stabilizes, to raise the effect of horticultural therapy program.
Plant fragrance is composed of highly volatile aromatic compounds, among which certain fragrance compounds have therapeutic effects. If these therapeutic effects are used by function, it will be helpful to increase the effect of horticultural therapy. Therefore, it is necessary to analyze the fragrance compounds of plants and to objectively verify the therapeutic effect of using them. In this study, we focused on the fragrance of these plants and investigated the content of functional aromatic compounds in plants with therapeutic effect and the way to increase them. In order to enhance the therapeutic effect of horticultural therapy using plants, the therapeutic effects of fragrance compounds were verified by stress relief, improvement of vascular function and changes of autonomic nervous system.
1. Analysis of Fragrance Components and Functional Fragrance Components of Plants In the horticultural therapy activity, the effect of the mental and physical stability and the stress reduction through the fragrance of plants, which is one of the functional elements of the plant, is expected to play a major role as another added value of plant. This study was conducted to select plants with high content 6 monoterpenes (α-pinene, β-pinene, 1,4-cineole, 1,8-cineole, limonene, linalool) that are known to have high stabilizing effects and apply them to horticultural therapy. For this purpose, monoterpene contents from five species of conifer (two species of Pinaceae, three species of Cupressaceae) and 4 Eucalyptus species (Eucalyptus cinerea, E. gunnii, E. polyanthemos and E. pulverulenta‘Babyblue’) which are widely used as green materials in flower design work and horticultural therapy activities were analyzed. As a result, the content of monoterpenes with therapeutic effect was the highest in the fir trees, and followed by pine trees and cypress trees with the difference between components. The content of linalool was highest in juniper trees. In Eucalyptus, Eucalyptus pulverulenta‘Babyblue’ contained the highest contents of α-pinene and 1,8-cineole.
2. Effects of Fragrance Components of Abies holophylla Max. Stress Relief, Vascular Function and Autonomic Nervous System The purpose of this study was to investigate the effects of fragrance of Abies holophylla Max., which is known to contain α-pinene, β-pinene, limonene, etc. on stress relief, vascular function and autonomic nervous system. In this cross-over design study, 15 healthy workers without otolaryngologic disease (37.6 ± 6.8 years) were randomized to either (1) plant fragrance inhalation or (2) odorless air inhalation by an experiment coordinator. Compared with odorless air inhalation, plant fragrance inhalation decreased brachial SBP from 117.64±4.89 to 108.36±3.92 mmHg (7.9%, p=.001), brachial DBP from 75.00±3.21 to 71.71±2.30 mmHg (4.4%, p=.004), central SBP from 104.86±4.22 to 97.21±3.28 mmHg (7.3%, p=.001), central DBP from 75.00±3.14 to 71.93±2.43 mmHg (4.1%, p=.015), and heart rate from 66.57±1.84 to 63.00±1.72 bpm (5.4%, p<.001). Arterial Stiffness Indexes such as AIx from 13.00±3.75 to 5.71±4.19% (56.1%, p<.001) and AIx@75 (7.71±3.51 to –0.29±3.93% (103.8%, p=.001) were also decreased significantly when fragrance was inhaled. There was a significant interaction effect between all treatments except diastolic blood pressure. Heart rate variability (HRV, 51.6%, p=.006) and flow-mediated dilation (FMD, 14.03%, p=.039) were significantly improved in plant fragrance inhalation vs. odorless air inhalation. In the stress response and recovery rate, the fragrance inhalation of the fir leaf alleviated the stress response in the arithmetic test and increased recovery rate. After stress, brachial SBP(Cont 14.3%, Treat 5.8%), brachial DBP(Cont 14.4%, Treat 9.4%), central SBP(Cont 14.7%, Treat 9.2%), and HR(Cont 14.6%, Treat 11.7%) were lower in fragrance inhalation(Treat) than those of odorless air inhalation(Cont) (p<.001), except for the central DBP(Cont 14.4%, Treat 14.7%). HRV (heart rate variability), which is an indicator of autonomic nervous system, was lowered in the LF/HF ratio (6.8%, p=.011) at the time of fragrance inhalation and the stress response was alleviated at the time of inhaling odorless air. Changes in vascular function were also less in AIx (Cont 34.7%, Treat 6.6%, p=.053) and AIx@75 (Cont 143.7%, Treat 89.9%, p=.008) than in the odorless inhalation. In addition, recovery rates were faster in all dependent variables except for cDBP.In conclusion, this study demonstrate that fragrance components of Abies holophylla Max. have positive effects of autonomic nervous system and improve vascular function. Therefore, it will be helpful for the selective use of plants to increase the treatment effect in horticultural therapy.
3. Effects of H2O2 and Salicylic Acid Treatment on the Monoterpene Content of Eucalyptus pulverulenta‘Babyblue’and Abies holophylla Maxim. This study was conducted to investigate the effect of hydrogen peroxide (H2O2) and salicylic acid (SA), treatment on the increase of monoterpene contents such as α-pinene, limonene and 1,8-cineole in Eucalyptus pulverulenta‘Babyblue’ and Abies holophylla Maxim. E. pulverulenta‘Babyblue’ shoots were soaked in various concentrations of chemical solution (0.3%, 0.5%, 1% H2O2 or 0.1 mM, 1 mM SA) or its leaves were sprayed with a 1% H2O2 or 0.1 mM SA solution. These were then analyzed for there α-pinene and 1,8-cineole contents at various periods after treatment (0, 0.5, 1, 2, and 4 h). A. holophylla Maxim. was soaked in 1% H2O2 or 0.1 mM SA solution then analyzed for the contents of α-pinene and limonene for 0, 0.5, 1, 2, and 4 hours after treatment, respectively. After 2 h of soaking, the α-pinene content in the 0.1 mM SA solution increased from 1.62 to 5.48 mg.L-1 (238%, p=.012), whereas the 1,8-cineole content in the 1 mM SA solution increased from 44.44 to 78.96 mg.L-1 (78%, p=.026) after 4 h of soaking. In response to spraying 0.1 mM SA on leaves, after 30 min, the α-pinene content increased from 1.62 to 3.91 mg.L-1 (141%, p=.007), whereas the 1,8-cineole content increased from 44.44 to 87.91 mg.L-1 (98%, p=.001), which means both increases were significant.
