Stat3, MAPK and PI3K/Akt signaling pathway which influence to cancer progression is triggered by binding IL-6 to IL-6R. The phosphorylation of stat3 is increased over 50% of breast cancer, and associated with cell growth, proliferation, migration and invasion of triple negative (TN) breast cancers. We firstly identified an IL-6 released from triple negative (TN) breast cancer cell line MDA-MB-231. MDA-MB-231 cell have known to be high invasive whereas MCF-7 cell is not invasive. IL-6 production was higher in invasive MDA-MB-231 than non-invasive MCF-7. Subsequently, we examined that MDA-MB-231 down-regulated IL-6 by treatment of IL-6 antibody or transfection IL-6 shRNA plasmid decreases the expression of pStat3, PI3K and pAkt, and it also induces the cell cycle arrest at G1 and G2 phase of MDA-MB-231 via down-regulation of cell cycle regulators (CDKs and Cyclins) by increasing p21 expression. Furthermore, sh-IL-6-transfected MDA-MB-231 cell is lower migration and invasiveness compared with sh-control MDA-MB-231 cell, as well as inhibited the expression of EMT factors, N-cadherin, snail, and vimentin. In vivo, the growth of tumors derived from sh-IL-6-transfected MDA-MB-231 cells was delayed compared with tumors derived from sh-control cell, and pStat3, pErk, PI3K, pAkt, snail, vimentin and N-cadherin is significantly lower in tumors derived from sh-IL-6-transfected cell. Secondary, we found the inhibitory effects of apigenin on cell growth and invasiveness of MDA-MB-231. Apigenin belongs to flavonoid is found in many vegetable, flower, and beverage such as parsley, thyme, chamomile and red wine. Many studies have reported various physiological activities of apigenin containing antioxidant, antibacterial, anti-inflammatory and anti-cancer effects. In recent study, the inhibitory effect of apigenin on several cytokines released from MDA-MB-231 was reported. Thus, we evaluated anti-cancer and anti-invasive effects of apigenin on MDA-MB-231 through a down-regulation of IL-6.
