In this thesis, I investigated the pathogenesis of the A/H1N1 2009 virus which contains the inhibitor resistant neuraminidase and is highly pathogenic in mice. I examined the immunogenicity and vaccine efficacy of A/H1N1 2009 split inactivated influenza A vaccine (GC1115) in mice and ferrets.
Following a brief description of the general influenza viruses in Chapter 1, in Chapter 2 covers the procedure to generate the mouse-adapted A/H1N1 virus as well as its virulence and pathogenic potential. The resulting A/California (CA)/04/09 virus harbors the H274Y mutation in the NA gene and the was adapted through continuous lung to lung passage in mice and its virulence and pathogenic potentials were evaluated. The mouse adapted H274Y (maCA04 H274Y) variants showed increased growth rate and virulence in vitro and in vivo while it maintained high neuraminidase inhibitor resistance. Although most of the maCA04 H274Y and maCA04 viruses contained mutations within HA (S183P and D222G) and NP (D101G), only the maCA04 H274Y virus showed a potential association with the K153E substitution in the HA gene. Given that the H274Y mutation confers decreased NA enzyme activity, it is meaningful that maCA04 H274Y viruses, but not maCA04 viruses, acquired a same K153E mutation in the HA gene and the K153E mutation affects receptor binding specificity by decreasing affinity for both α-2-3 and α-2-6-sialic receptors which make to restore the balance between HA and NA activities. In Chapter 3, To evaluate the vaccine efficacy of A/H1N1 2009 split inactivated influenza A vaccine (GC1115), we demonstrated the immunogenicity and protective efficacy of GC1115 in mice and ferret models. Therefore non-adjuvanted inactivated split virus influenza vaccine has the merit of developing without safety issues of new adjuvant on urgent pandemic situation. The immunogenicity of GC1115 was confirmed after intramuscular administration of 1.875, 3.75, 7.5, and 15 µg hemagglutinin antigen (HA) in mice and 7.5, 15, and 30 µg HA in ferrets with 3-week intervals. The mice of mock (PBS) and 1.875 ug HA/dose immunizing groups were succumbed to data in 13 days by following the A/California/04/09 infections, but all mice from more than 3.75 ug HA/dose groups were 100% protected from lethal challenge of the virus. In ferrets, although 15 and 30 µg HA immunizing groups induced the detectable HI antibodies even in a single dose, all double immunization groups were seroconverted with more than 80 HI unites. Following the A/California/04/09 challenges, GC1115 immunized ferrets showed attenuated clinical symptoms only in short period (3-4 days). Further, the GC1115 immunized ferrets displayed significantly lower viral titers in the upper respiratory tract (nasal cavity).
