IFA 결과 CB2 페럿 항 혈청은 CB1 (유전자형 B)와 CB2 (유전자형 A) 바이러스를 모두 검출 할 수 있음을 확인하였으며, 동종의 유전자형인 유전자형 A 바이러스에서 더 강한 형광이 관찰되었다. To develop the large scale serological diagnostic assay for SFTSV infection, we evaluated two different enzyme-linked immunosorbent assays (ELISA) using nucleocapsid (N) protein and glycoprotein N (Gn) proteins of SFTSV CB1 strain and compared their sensitivity and specificity with Immunofluorescence assay (IFA). The recombinant N protein (rN) –based ELISA test showed more sensitivity and broad cross-reactivaty between two different genotype A and B strains compared with those of recombinant Gn protein (rGn) –based ELISA tests. However, rGn-based ELISA showed more specificity to homologous genotype. The IFA results showed that CB2 ferret anti-sera could detect both CB1 (genotype B) and CB2 (genotype A) viruses although the higher intensity of IFA was observed in genotype A strains. Further, Western blot assay showed that purified CB1 and CB2 viruses have different pattern of positive bands against Gn/glycoprotein C (Gc) proteins although the N specific bands were detected in both antibodies suggesting antigenic differences between genotype A and B SFTSV strains. To evaluate the recombinant N and rGn-based ELSIA assay for clinical applications, we conducted sero-prevalence studies using serum specimens of domestic deer and wild migratory birds. The rN-based ELISA assay revealed that about 14.0% (30 of 215) and the rGn-based ELISA has 7.9% (17 of 215) of domestic deer sera were positive for SFTSV antibody. Further, more than 32.2% (127 of 394) sera of wild migratory birds were positive for rN-based ELISA and 17.8% (70 of 394) positive for rGn-based ELISA. Therefore, SFTSV antibody suggesting high incidence of SFTSV infection in natural environments. Taken together, in this study, we develop the rN- and rGn-based ELISA tests and these methods could be applicable for general sero-prevalence studies of SFTSV infections, while rGn-based ELISA could be applicable for genotype sero-prevalence study of certain specific genotype.
