Our previous study had identified the SNP (g.81018510 T > C) and indel (g.81018497 D > I) located in the promoter of APM1 to have a significant effect on marbling in Hanwoo. APM1 encodes an adipocytokine called adiponectin, which plays a significant role in lipogenesis. The aim of this study was to verify and validate the effect of the SNP and indel on marbling and other carcass traits in a large, representative, countrywide population of Hanwoo cattle. The economic traits measured were carcass weight (CAW), loin eye area (LEA), backfat thickness (BFT), marbling (MAR), maturity(MAT), meat color(MCO), fat color(FCO) and texture(TXT).
Primers were designed to amplify 346 bp of the genomic segment that contained the targeted SNP (g.81018510 T>C) and the indel (g.81018497 D>I). After data curation, the genotypes of 4,458 individuals identified using direct sequencing analysis estimated frequencies for C (0.549) and T (0.451) respectively showing genotype frequencies for CC (0.303), CT (0.491) and TT (0.206). The genotypes were significantly associated with CAW, LEA and MAR, MAT, TXT. The indel of 6,944 individuals had significant effect on MAR and MAT. The allele frequencies was estimated at (DEL, 0.863) and insertion (INS, 0.137) presenting genotypes of D/D (0.756), D/I (0.215), and I/I (0.029). Significant departure from Hardy-Weinberg equilibrium was not detected for both the SNP and the indel. The haplotype were significantly associated with CAW(p = 0.04) and MAT(p <.001)
The SNP genotypes showed significant association with CAW, LEA and MAR, MAT, TXT additive genetic effects, while the indel was significantly associated with MAR and MAT. The results confirmed that the variants can be used as a genetic marker for improving traits in Hanwoo.
