FG340과 Pf04 두 균주를 Petri-dish (90 × 15 mm)조건에서 12가지 곡물에 고체 배양을 진행한 결과, 기장, 보리, 쌀, 조 에서 1 × 109conidia/g이상의 포자 생산성을 확인할 수 있었다. 또한 곡물배지인 조에서 배양되어 수확된 포자에서 열안정성이 가장 높은 것으로 확인되었다. 수확된 포자를 현장적에서 경엽 및 관주 살포용으로 적용하기 위하여, 포자 현탁액을 효과적으로 제조할 수 있는 계면활성제를 선발하였다 (고체배양 산물에서 포자를 용출시키기 위한 용도의 계면활성제). FG340과 Pf04 균주는 UV-C 자외선에 노출되었을 때, 일부 열안정성이 향상된 자연 변이주가 유도되었다. 본 연구를 요약하면, 첫째, 4가지 해충에 대한 I. javanica FG340과 Pf04의 살충성 검정을 통해 농업 해충 방제에 효과적으로 적용이 가능한 소재임을 확인하였으며, 둘째, 포자 생산성 및 열안정성을 향상시킬 수 있는 곡물 기질인 조와 기장을 선발하였으며, 마지막으로 UV-C와 같은 환경 조건 변화를 통하여 열안정성이 향상된 균주 개량이 가능함을 확인하였다. 앞으로 추가적인 현장적용 기반연구를 통하여 본 연구의 산업화가 이뤄질 수 있도록 노력할 예정이다.

However, optimizing conidial production systems to support high productivity and conidial stability still needs additional efforts for successful field application and industrialization. Although many virulent entomopathogenic fungal isolates have been investigated as potential candidates in a laboratory environment, very few of the isolates are being used in practice for application in agricultural fields as commercial products. Isaria javanica is an entomopathogenic fungus that is pathogenic to diverse coleopteran and lepidopteran insects and thought to be a good candidate as biopesticides. In this work, the basic characteristics of two entomopathogenic I. javanica FG340 and Pf04, were investigated in morphological features, genetic identification, and virulence against diverse coleopteran and lepidopteran insects, such as Thrips palmi, Riptortus pedestris, Bemisia tabaci, and Tenebrio molitor. The feasibility of grains substrates for conidial production was assessed, particularly focusing on conidial productivity and thermotolerance. Moreover, thermotolerance of the Isaria isolates was improved by exposing the isolates to environmental stress. I. javanica FG340 and Pf04 conidia were solid-cultured on 12 grains for 14 days in Petri dish conditions. Of the tested substrates, Italian millet, perilla seed, millet and barley-based cultures showed high conidial production. The four-grain substrates produced more than 1 × 109 conidia/g of substrates in I. javanica FG340 and Pf04. Both of two isolates had enhanced thermotolerance when cultured on Italian millet. In application, it was easy to make a conidial suspension using the cultured grains. And several surfactants were tested to release conidia from the cultured grain substrates. In addition, the two I. javanica isolates showed high thermotolerance when exposed to UV-C radiation. In summary, first the fungal thermotolerant conidia can be produced by the two methods, grain-based solid culture system and exposure to UV-C radiation. Secondly the I. javanica showed high virulence against diverse coleopteran and lepidopteran insects. Lastly this work suggests several possible inexpensive grain substrates by which to promote conidial production combined with enhanced stability against exposure to high temperature.
