Lipophagy, a catabolic process with an essential function in maintaining cellular and tissue homeostasis, is an alternative pathway of lipid metabolism through the lysosomal degradative pathway of autophagy. The ability of autophagy to contribute to maintaining lipid homeostasis is particularly relevant in the context of genetic lysosomal storage disorders where perturbations of autophagic flux have been suggested to contribute to non-alcoholic fatty liver disease (NAFLD). Fenofibrate, a peroxisome proliferator-activated receptor α (PPARα) agonist, controls lipid metabolism by enhancing the expression of genes involved in fatty acid uptake and mitochondrial fatty acid oxidation, which are widely used in treating hyperlipidemia. However, the effect of fenofibrate on amelioration of fatty liver disease through the regulation of lysosomal-lipophagy remains largely elusive. In this study, we first found that fenofibrate treatment induces autophagic flux in both hepatocytes and mouse fatty liver tissue. Moreover, fenofibrate treatment promotes lysosomal function as evidenced by increased lysosomal acidification and lipophagy activity. Second, fenofibrate can induce lipophagy activation independently of mTORC1. Fenofibrate failed to inhibit mTORC1 and continued to activate lysosomal function in tsc2-/- MEFs with mTORC1-hyperactive conditions, indicating that fenofibrate-treated lysosomal activation is achieved independently of mTORC1. Third, fenofibrate treatment induces transcription factor EB (TFEB) and transcription factor E3 (TFE3), a master key regulator of autophagy and lysosomal biogenesis and function, based on the following observations: a) fenofibrate directly induces TFEB/TFE3 expression, b) TFEB/TFE3 nuclear translocation, c) increased transcriptional activation of TFEB/TFE3, and d) activation of AMPK-CaMKKβ signaling through upregulation TFEB/TFE3-mediated lipophagy. Intracellular Ca2+ is one of the regulators of autophagy, we analyzed the possibility that fenofibrate promotes between lysosomal calcium signaling such as MCOLN1, calcineurin, and TFEB/TFE3. Finally, we found that fenofibrate induces lysosomal Ca2+ release through MCOLN1 and activates calcineurin, thus promoting TFEB/TFE3 nuclear translocation. Finally, activation of lipophagy and lysosomal biogenesis result in amelioration of lipid accumulation and fatty liver injury in the fenofibrate-treated liver, indicating that lipophagy and lysosomal activation serves to ameliorate hepatic fat accumulation in NAFLD. Taken together, our study provides a novel insight into the regulatory mechanism of fenofibrate on lipophagy and lysosomal degradation, which may facilitate establishment of fenofibrate as a potential NAFLD therapeutic.
