Now a days eDNA (environmental DNA) metabarcoding is used as a sophisticated technique to assess the fish diversity of the marine environment. This method is based on DNA analysis and allows accurate identification, and can detect species with a low frequency of occurrence that is not caught by fishing. The objective of this study was to investigate fish fauna in the coastal area of the eastern part of the south sea of Korea using underwater visual census (UVC) and eDNA analysis. To observe fish fauna in the eastern part of the south sea of Korea (Tongyeong, Busan, and Ulsan), monthly UVC were conducted for one year from November 2016 to October 2017, June to October 2018. Samples for eDNA analysis were collected for five months from June to October, 2018. A total of 14,421 individuals of 68 species were found in Tongyeong coast, 2,191 individuals of 36 species in Busan coast and 4,534 individuals of 39 species in Ulsan coast using UVC during the survey period. Tongyeong, Busan and Ulsan were dominated by Chromis notatus, Halichoeres tenuispinis and Siganus fuscescens which were 25.3%, 19.1% 32.4% of the total number of individual respectively. eDNA analysis detected 58 species in Tongyeong, 103 species in Busan, 102 species in Ulsan, and combinedly 145 species all location. Although eDNA metabarcoding is still not using sufficiently in different environmental condition but because of it’s specialized characters such as non-destructiveness, detection of fish fauna quickly and easily, it will be a standard tool for surveying fish communities.
