Streptococcus iniae is a gram-positive, non-Lancefield streptococci, sphere-shaped bacterium. S. iniae causes serious disease in a wide range of fish species. It is one of the major pathogens that damage aquaculture and fishery industry. Especially in a narrow and dense aquaculture, the disease spreads quickly and causes enormous damage. S. iniae has many virulence factors such as hemolysin (Streptolysin S). And some S. iniae have a toxin-antitoxin gene. In Streptococcus pneumoniae, virulence is known to be variable depending on the presence or absence of these genes. Toxin antitoxin (TA) system is a set of two or more closely linked genes that together encode both a protein 'toxin' and a corresponding 'antitoxin'. TA system is a regulatory gene found in bacteria, and exists as an operon. Toxin is an enzyme that targets various targets in bacterial cells (DNA replication, tRNA synthesis, Cell wall synthesis ...). TA system has various types including Type I to Type VI. Its classification is divided into interactions (RNA-RNA, RNA-protein, protein-protein ... etc). Among them, the type II TA system is protein-protein interaction which produces a labile antitoxin protein and a stable toxin protein. Antitoxin binds toxin to prevent toxin from activating. In particular, toxin acting on the cell wall is known to be activated by stress factors such as antibiotics, osmotic shock, and long term incubation, and viability decreases. The ε/ζ TA system has been reported to affect virulence and viability in most cases of Type II TA system found in Gram-positive bacteria. In this study, I predicted that the ε/ζ TA system on the plasmid of Streptococcus iniae FP5228 would affect the virulence and cell viability. To confirm this, I constructed Toxin ζ overexpression model in E. coli and TA system deletion strain by curing plasmid of S. iniae FP5228. As a result of overexpression of toxin, the number of viable cells decreased and the morphology of the cells was changed. This implies that Toxin ζ affects the viability of bacteria and can cause morphological changes through cell wall weakening. In S. iniae without TA system, it was confirmed that the decrease of viable count was alleviated. In addition, the results of the virulence test and the fish model of the cell showed that the TA system showed lower hospital in the strain removed. It is shown that the TA system of S. iniae affects viable count and virulence. Through this study, it was suggested that the removal of TA system of S. iniae may be helpful for continuous research and development of attenuated strains. Through this study, it was suggested that the removal of the TA system of S. iniae FP5228 may help to increase stability in long term studies and may help to develop attenuated strains.
