Adipose tissue is identified as an abundant source of mesenchymal stem cells (MSCs) which is a promising candidate in stem cell transplantation. For clinical applications, adipose-derived mesenchymal stem cells (ADMSCs) are stored in optimal storage solutions to be readily available before transplantation. However, the effects of these temporary solutions on MSCs remain unknown or merely discussed. In this study, I aimed to find out widely used stem cell carrier by searching for the last 10 years to examine the effects of storage solutions on the viability and potency of MSCs prior to cell transplantation. According to the search findings, the freshly harvested canine ADMSCs were suspended in five different solutions namely 0.9% saline (saline), phosphate-buffered saline (PBS), 5% dextrose solution (5% DS), heparin in saline (Hepa-Sal) and Hartmann solution incubated for 1, 3, 6 and 12 hours (h) at 4°C. Cell viability, confluence ability, proliferation, self-renewal ability, differentiation capability and the gene expression level were investigated. When ADMSCs were stored in parenteral solutions, it showed progressive deterioration in their viability, attachment ability and the proliferation capability. Osteogenic and chondrogenic differentiation ability were also affected over time. ADMSCs in saline stored for 12 h showed the highest survival viability and confluence rate in 72 h. On the other hand, ADMSCs maintained the proliferation when stored in 5% DS for 12 h. ADMSCs in Hepa-Sal and Hartmann solutions showed the superior chondrogenic and osteogenic ability respectively. The expression level of stemness marker Sox2, MSCs marker CD90 and CD105 were also reduced with time. My outcome suggests that researchers should take into account to store stem cells in Food and Drug Administration (FDA) approved solutions for high viability and efficacy (saline, 5% DS, Hepa-sal and Hartmann solution) rather choosing PBS that has no FDA approval.
