Over the past few decades, identifying the complex mechanisms of brain disease has emerged as an important issue. The brain disease is influenced by complicated biological processes such as genetic polymorphisms, molecular process, neural pathway, and behavioral process. Traditionally, researchers have tried to uncover the mechanisms of the disease by analyzing a bio-signal from a single modality (e.g., genomics, proteomics, molecular biology, radiology, and neuroscience). However, it is difficult to fully explore the disease mechanism by analyzing a single source of biomedical information. In recent years, a research field known as imaging genetics has been introduced where we can investigate relationships between genetic polymorphisms and neuroimaging features (e.g., atrophy, cortical thickness, and connectivity, etc.). Diverse brain phenotypes from neuroimaging and genetic variation could jointly contribute to complex diseased related phenomena. Thus, combining genetic and imaging information might help us to identify or recognize patterns of biomedical signals, which might improve our understanding of the disease and lead to developing effective therapies. In this dissertation, we developed an effective multivariate imaging genetics analysis model combining multi-modal neuroimaging data with genetic information. The connectivity-based regularization method was developed to encourage solutions that are not only mathematically well-defined but also biologically meaningful. Then, we confirmed the feasibility of our proposed imaging genetics method and its clinical efficacy using real datasets of Parkinson’s disease. Multivariate models have been accepted as critical tools to investigate the significant associations between imaging features and genetic markers in imaging genetics research. Current multivariate models still have shortcomings related to overfitting problem and clinical reliability. In this dissertation, the connectivity-based regularization method has been proposed to overcome these problems. We hope this dissertation will provide a foothold for the development of advanced imaging genetics models to improve our understanding of various brain diseases.
