Rat sarcoma (RAS)-extracellular signal-regulated kinase (ERK) signaling is a ubiquitous signaling pathway and involved in numerous cellular functions and biological processes such as cell proliferation, migration, fate specificity, and differentiation. RAS-ERK signaling also plays critical roles in brain functions such as learning and memory by regulating the neurotransmitter release and transcription. RAS-ERK signaling consists of the major components such as RAS, RAF, MEK, and ERK and supporting modulators such as SHP2, NF1, and SynGAP1. RASopathies are caused by mutations in the genes that encode components of the RAS-ERK signaling pathway. RASopathies share clinical symptoms, including heart defects, delayed growth, and intellectual disabilities, while each RASopathy also displays unique phenotypes. Recent studies using RASopathy mouse models showed that distinct cell types in the nervous system are differentially affected among different RASopathies, such as Noonan syndrome (NS) and neurofibromatosis 1 (NF-1). However, the molecular mechanisms underlying the cell type-specific pathophysiology are still unclear. In my dissertation, I hypothesized that each cell type might contain distinct RAS-ERK signaling network contributing to the cell type-specific pathophysiology in RASopathy. In chapter 1, I reviewed the cell type-specific roles of the RAS-ERK related genes in learning and memory. Furthermore, I reviewed previous literatures investigating the molecular mechanisms underlying the cell type-specific pathophysiology in RASopathy by summarizing and analyzing the cell type-specific phenotypes in various animal models of RASopathies. In the following chapters, I investigated molecular mechanisms that could account for the cell type-specific effects of RAS-ERK signaling associated genes using single cell type-transcriptome analyses together with biochemical, behavioral, and electrophysiological approaches. In chapter 2, cell type-specific gene expression profiles in mouse hippocampus were analyzed to examine whether the RAS-ERK signaling networks are different between excitatory and inhibitory neurons. I revealed that the compositions of RAS-ERK signaling pathways are different between two neuronal cell types in mice. For example, Nf1 expression is significantly higher in inhibitory neurons than in excitatory neurons in mouse hippocampus. In consistent with transcriptome data of mice hippocampus, I confirmed the expression level of NF1 mRNA is also higher in inhibitory neurons than in excitatory neurons in the human cortex. In chapter 3, I tested the effect of expressing a NS-associated SHP2D61G in excitatory or inhibitory neurons on synaptic plasticity and behavior. SHP2D61G expression resulted in the excitatory neuron-specific phenotypes in ERK activation, long-term potentiation and memory. I showed that excitatory neuron-specific disruption of the interaction between SHP2 and its binding protein Gab1 can rescue the deficits in LTP. Finally, I showed that SHP2D61G can activate Ras signaling pathway in inhibitory neurons by expressing SHP2-interacting proteins Gab1 and Grb2 in inhibitory neurons. In chapter 4, I investigated the impacts of inhibitory neuron-specific manipulation of RAS signaling by expressing KrasG12V in hippocampal inhibitory neurons. I found that the expression of KRASG12V in inhibitory neurons impairs learning and memory by increasing RAS-ERK signaling in inhibitory neurons. In my dissertation, I demonstrated that RASopathy-associated genes are differentially expressed between excitatory and inhibitory neurons, which may be responsible for the cell type-specific pathophysiology of individual RASopathy. Furthermore, my study suggests that when developing drugs for cognitive dysfunction of RASopathy patients, cell type-specific approach should be considered to increase drug specificities.
