Background: Dermal fillers are of interest due to simple, short surgical procedures and rapid facial rejuvenation. There are various types of dermal fillers (HA, PLLA, PDO, PCL, PLA). Among which the most common types of fillers include HA (hyaluronic acid), but small fragments of hyaluronic acid cause inflammation and have side effects such as erythema, mild edema, hematoma, itching, and pain. Therefore, the filler development of other materials is active in order to reduce such side effects. It is well known that PN is DNA-based material extracted from salmon and has excellent biocompatibility and induces tissue regeneration. Objective: To compare the efficacy and safety of HA fillers and novel Hyaluronic acid-Polynucleotide (HA-PN) complex fillers. Methods: The characteristics of HA filler and HA-PN complex filler were compared by SEM and Rheometer. Also, cell migration by cell scratch assay and collagen synthesis was compared between human and mouse fibroblast cells. HA filler and HA-PN complex filler were injected with each filler in SKH-1 hairless mice, to determine the changes in volume up to 24 weeks and analyzed safety and the amount of collagen synthesis, and TRPV4 protein expression in histological evaluation. Results: There was no morphological difference between HA filler and HA-PN complex filler, but HA-PN complex filler had higher viscosity and elasticity. In addition, the HA-PN complex filler has excellent cell migration and collagen synthesis. In animal models, safety and durability are similar to HA fillers and less stimulation by filler injection. Conclusion: These findings suggested that HA-PN complex fillers can stimulate fibroblast growth and can benefit from volume growth and skin regeneration. This is the first study to demonstrate the durability, efficacy and safety of HA-PN complex fillers.
