The epidermal growth factor receptor (EGFR), a member of ErBb family, plays a crucial role in activation of a plethora of key cellular signaling programs leading to proliferation, differentiation and survival. Consequently, its aberrant activation is closely associated with cancer pathogenesis. Recently, our group has reported a novel molecular mechanism of phosphorylation-mediated feedback inhibitory role of Mig6 (mitogen-inducible gene-6) against EGFR activation. In addition, we showed high frequency of hemizygous focal deletion of Mig6 in EGFR-amplified glioma patient samples. In this study, we sought to explore the functional significance of Mig6 genomic alteration in tumorigenesis. To this end, we characterized 15 recurrent Mig6 mutations identified from various cancer with in vitro models. We found that a subset of mutant Mig6 are not able to bind to EGFR and failed to inhibit EGFR activation. In addition, these Mig6 mutants did not suppress the growth of GBM cells addicted to EGFR or ERBB2 mutants. Based on these results, we propose that aberrantly activated EGFR signaling pathways caused by loss-of-function Mig6 mutations may lead to cancer development and Mig6 mutation status may be key genomic determinants for EGFR-targeted therapy in cancer patients.
