S. aureus MW2 감염에 대한 세균-반응성 단일클론항체의 효과를 확인하기 위하여, S. aureus MW2를 마우스의 정맥내에 투여하였고, 세균-반응성 단일클론항체를 정맥내에 투여하였다. 연구결과: CpG-DNA의 투여로 S. aureus MW2 감염에 대한 마우스의 생존율이 증가하였고 마우스에서 세균의 제거가 증가되었다. Background: CpG-DNA activates various immune responses, contributing to the host defense against bacterial infection. Several mechanisms involved in the function of CpG-DNA against bacterial infection were reported. The presence of bacteria-reactive antibodies in normal serum was also reported before. However, relevance of CpG-DNA with bacteria-reactive antibodies was never investigated. Here, the biological functions of CpG-DNA regarding production of bacteria-reactive antibodies were investigated. Methods: To investigate the effects of CpG-DNA on methicillin-resistant Staphylococcus aureus (S. aureus) MW2 infection, BALB/c mice were intraperitoneally (i.p.) injected with CpG-DNA, and intravenously (i.v.) injected with S. aureus MW2. To determine the production of bacteria-reactive antibodies, ELISA was performed using poly-L-lysine coated plates. To determine the influence of antibodies on phagocytosis in the mouse peritoneal cavity, the mice were i.p. injected with FITC-labeled S. aureus MW2. After 1 h, the peritoneal cells were harvested and stained with cell-specific markers. To determine the effects of the bacteria-reactive monoclonal antibody on Saureus MW2 infection, the mice were i.v. injected with S. aureus MW2, and immediately injected with the bacteria-reactive monoclonal antibody. Results: Administration of CpG-DNA increased survival of mice against infection with S. aureus MW2 and facilitated bacterial clearance in mice. While infection of S. aureus MW2 induced drastic decrease of total cell numbers in the peritoneal cavity, bone marrow, and spleen, pretreatment of the mice with CpG-DNA commonly protected decrease of immune cell populations. Injection of mice with CpG-DNA induced increase of bacteria-reactive antibodies, which can bind to diverse species of bacteria, in the peritoneal cavity and serum through TLR9-dependent pathway. The bacteria-reactive antibodies were produced in both B1 and B2 cells of peritoneal cavity in response to CpG-DNA, and the antibodies enhanced phagocytosis in the peritoneal cavity of mice. A hybridoma clone producing bacteria-reactive 3F5H6 mIgG monoclonal antibody was selected from CpG-DNA stimulated-peritoneal B cells. The purified bacteria-reactive 3F5H6 mIgG monoclonal antibody was reactive to various bacteria and enhanced phagocytosis of S. aureus MW2 in a macrophage cell line and the primary peritoneal cavity cells. Injection of mice with the bacteria-reactive monoclonal antibody showed therapeutic effects against infection of S. aureus MW2 with enhanced survival rate and bacterial clearance. Conclusions: These results suggest that CpG-DNA enhances the antibacterial activity of the immune system by protecting immune cells and triggering the production of bacteria-reactive antibodies. Consequently, these results support that bacteria-reactive monoclonal antibodies could aid in the treatment of antibiotic-resistant bacterial infections.
