ToxCast database contains 9,076 chemical toxicity data for 1,192 bioassays. However, there are limitations to using toxicity data on individual biological targets in the toxicity assessment of chemicals. Humans are exposed to numerous environmental chemicals such as particles, fibers, nanomaterials, gases, and volatile organic chemicals through inhalation pathways. However, the traditional toxicity assessment of chemicals causing inhalation toxicity are not only very expensive, but also technically difficult and time consuming. In order to efficiently and rapidly screen the potential inhalation toxicity of a large number of chemicals, the AOP for inhalation toxicity screening is required. The overall aims of this project were to 1) develop AOPN for inhalation toxicity screening; and 2) explore the availability of some computational approaches in the development of AOP and propose an inhalation toxicity screening Integrated Testing Strategy (ITS). In order to achieve these aims, the following studies were conducted. First, development of AOPN on pulmonary fibrosis was conducted. The AOP was developed using information from the literature related to the mechanism of pulmonary fibrosis. For an overall assessment of the AOP, a literature review was conducted and the AOP was evaluated based on the Bradford-Hill considerations. The overall Biological Plausibility and Essentiality of each KE are high, but the Empirical support and quantitative relationships are moderate or weak. Second, identification of potential chemicals for AOP validation was conducted using ToxCast databases and deep learning models. Chemicals bearing potential inhalation hazard and exposure hazards were identified from the database. Then the systematic analysis was conducted to identify potential chemicals involved in this AOP, using the ToxCast database and deep learning artificial neural network models. The activity of chemicals do not tested in ToxCast were then predicted using these deep learning models. Both approaches identified a set of chemicals that could be used to validate the AOP. Third, MIE validation was conducted using in silico molecular docking and in vitro assay. On the potential chemicals, MIE was validated first with in silico approach using molecular docking. Among the chemicals having low binding energy to PPARγ receptor, rotenone, triorthocresyl phosphate, and castor oil were selected as PPARγ antagonist for validation of MIE of this AOP. These results suggested that in silico molecular docking approach has a potential to be applied for identifying MIEs. Fourth, the causal relationship between KEs was identified using Bayesian Network model. The in vitro test data on the biomarkers related to fibrosis and cell death were collected from the previous studies. Based on these data, AOP of stress response leading to fibrosis was developed, and BN model was learned for identifying probabilistic causal relationships between the KEs. The effect of KE was altered by the exposure of MWCNTs. Overall, our approach shows that BN modeling has the potential to identify causal relationships in an AOP. Lastly, we applied AOP approach in a case study to identify the toxicity pathways of environmental chemicals, such as, PM2.5. Mechanism of toxicity of individual chemicals in PM2.5 were systematically identified. Using both the ToxCast database and deep learning models, active chemicals on each biological target were identified. Through molecular docking and in vitro PPARγ activity assay, potential for pulmonary fibrosis of DPM were identified. This study suggested that in silico assay combined with experimental validation is a useful and efficient tool for predicting environmental chemicals. Based on these studies, we proposed Integrated Testing Strategy (ITS) for screening inhalation toxicants based on database and developed inhalation toxicity AOP. The computational approaches, used in this study, make it possible to utilize data from a variety of sources and types, especially those produced by different methods. This is a suitable approach for AOP aimed at maximizing the use of existing knowledge. The integration of AOP with rapidly developing computer technologies will be a new direction to accelerate mechanism-based chemical toxicity assessment.
