In this study, we investigated the antioxidant and antimicrobial activities of olive desalted in brine olives and olive leaf extracts. And functional dairy products were developed by measuring the physicochemical characteristics and antioxidant effects of yogurt with desalted green olive and yogurt with olive leaf extract. First, this study examined the effect of desalting brined olives (green, black) for 6 h on the total phenolic content (TPC), antioxidant activity, and antimicrobial activity. The TPC values decreased in the order salted green olive (SGO) > salted black olive (SBO) > desalted green olive (DGO) > desalted black olive (DBO), but there was no significant difference between DGO and SBO (p>0.05). The DPPH radical scavenging activities of SGO and DGO were the highest and showed no significant difference (p>0.05). Although desalting has low effect, the antioxidant and antimicrobial activities of the desalted samples remain high. Second, results of this study demonstrated that green olive powder might be used to improve the antioxidant capacity and sensory characteristics of yogurt. The more time of titratable acidity went by, the more it increased. And the more time of syneresis went by, the more it increased, but GY3 of them showed the lowest syneresis. The antioxidant activity of GY5 was found to be the highest among the four groups at day 1 of storage. In the sensory evaluation sweet and overall of GY3 indicated the highest score among the four groups. Third, we investigated the influence of ethanol, methanol, acetone (50%, 70%, and 90% v/v), and distilled water on selected properties of olive leaf extract and determined the yield, total phenolic content (TPC), antioxidant activity, and antimicrobial activity. Olive leaf extracts using 90% methanol had high levels of polyphenols and were highly antioxidant and antimicrobial. HPLC analysis showed that the highest oleuropein contents corresponded to the extracts obtained using 90% methanol. Fourth, the objective of this study was to determine the antioxidant and quality characteristics of yogurt containing olive leaf hot water extract stored at 4°С for 15 days. After storage, the lactic acid bacteria count showed no significant difference between yogurt samples either in the presence or absence of olive leaf extract (p>0.05). The total phenolic content, DPPH radical scavenging activity, and reducing power increased upon increasing the concentration of the olive leaf hot water extract. In conclusion, it was shown that desalted green olive powder can be used to improve the antioxidant capacity and sensory properties of dairy products. The results of the olive leaf extract enable the commercial application of natural extracts with antioxidant and antimicrobial activitioes, and proved that the manufacturing of yogurt containing olive leaf extract was beneficial to health.
