Neural stem cells (NSCs) are defined as stem cells able to self-renew and generate all cell types of the nervous system. It is important to understand the underlying mechanisms that determine cell fate to efficiently modulate neurogenesis. MicroRNAs are a class of small non-coding RNAs of 19-25 nucleotides that control gene expression at the post-transcriptional level by blocking messenger RNA (mRNA) translation or degrading mRNA. As small RNA regulators, microRNAs are involved in numerous cellular processes including proliferation and differentiation and can regulate neuronal function, maturation, and development by controlling target genes. An important concept in microRNA research of the nervous system is that microRNAs play a critical role in regulating the balance between self-renewal and cell fate determination of neural stem cells. Cell- or tissue-specific microRNAs play an important role in identifying of specific microRNAs function. Therefore, there is a need for studies to investigate the role of specific microRNAs in undifferentiated and differentiated cells by screening for microRNAs specifically expressed in NSCs. In this study, NSC-specific microRNAs were identified by qPCR array based screening, their specific expression in NSC-derived glial and neuronal cells was confirmed, and the role of specific microRNAs in NSCs and in differentiation into glial and neuronal cells derived from NSCs in vitro was examined. Chapter 1 explains the general contents and purpose of this dissertation, as well as research trends regarding the role of microRNAs in NSCs, nervous system, and cell fate determination. Chapter 2 describes the establishment of an efficient differentiation method to obtain NSC-derived glial cells for the study of the role of microRNAs in NSCs and glial cells derived from NSCs. Chapters 3 and 4 describe the role of NSC-specific microRNAs in NSCs, NSC-derived glial cells, and NSC-derived neuronal cells. MicroRNAs specific for NSCs were found to be expressed at similar or higher levels in glial and neuronal cells in comparison with their levels in NSCs. To verify the role of these microRNAs, an inducible microRNA sponge system was integrated into the cells to induce downregulation of specific microRNAs. A non-infected NSC line and a scrambled vector-infected NSC line were used as controls. Downregulation of a microRNA that is specific for NSCs is expressed at a higher level in astrocytes than in NSCs was found to induce apoptosis of NSCs and astrocytes, and to increase the expression of the target gene. In contrast, downregulation of a microRNA specific for NSCs and highly expressed in neuronal cells had no effect on the characteristics of NSCs, although this downregulation is expected to target genes involved in neuronal differentiation and maturation. Overall, this study established an efficient method to differentiate NSCs into glial cells and identified the expression patterns of microRNAs specific to NSCs, glial cells, and neuronal cells, and the roles of these microRNAs in undifferentiated and differentiated cells. These findings suggest the possibility of microRNAs used as biomarkers in neurological diseases and that specific microRNAs can be used in brain disease treatment.
