Hence, I examined the role of the LHb in conditioned inhibition using the extinction procedure, one of the traditional learning procedures to measure conditioned inhibition. The first experiment was conducted to examine whether the LHb is involved in conditioned inhibition by the measuring c-Fos levels of the LHb after the extinction test or not. Expression levels of c-Fos were increased in the LHb of the animals with the presentations of CS alone after the excitatory conditioning. On the contrary, c-Fos levels were increased in the VTA and SNc, but not LHb, of the animals that received the continued paired learning. The second experiment examined the effects of the chemical lesion on the extinction of conditioned responses to clarify the involvement of the LHb in extinction. The LHb lesions delayed the extinction rates of conditioned response. Unlike the second experiment, the third experiment examined chemogenetic activation effects of the LHb by designer receptors exclusively activated by designer drugs (DREADDs) on the extinction rates of conditioned response. The chemogenetic activation of the LHb facilitated the extinction rates. Furthermore, the c-Fos-positive signals in animals with the presentation of CS alone were mostly observed in the medial part of LHb. These results indicate that the LHb is a critical neural substrate of the acquisition of conditioned inhibition. Both conditioned excitation and inhibition are the fundamental learning process to guide adaptive behavior. Involvements of LHb and its interaction with the midbrain dopaminergic neuronal activity in the associative process of conditioned inhibition would contribute to the understanding of the current conception of conditioned inhibition.
