However, the lack of sensitivity, wide selectivity, and difficulty in development due to the limited number of genetic systems make hard to apply biosensors for practical monitoring in environmental systems. In order to solve low sensitivity, we used mutant E. coli strain that lost a gene named as copA involved in copper transporting system. Removal of the copA encoding copper transporting protein caused the internal accumulation of copper ions in E. coli by interfering the copper homeostasis control system. In addition, the amount of cueR which is transcriptional regulator known to interact with copper ions was critical to optimize sensitivity. Secondly, the metal binding site (MBL) of the regulatory protein arsR which response to arsenic and antimony was engineered. The antimony-response biosensor was engineered by minimizing the binding of arsenic ions and increasing the binding of antimony ion by removing the 37 cysteine and relocating the position. Third, in order to solve the limitation of biosensor development due to the limitation of the operon's diversity, the reporter gene eGFP was engineered to eliminate the use of the stress-response operon in the manufacturing stage and to bind to the metal using the reporter gene. MBL which binds to cadmium and mercury based on the regulatory protein zntR, was inserted into the separated loop After separating the amino acid regions 1-9 and 10-11 of eGFP, As a result, when cadmium ions were combined to MBL, the separated fragment of eGFP was closer and showed a fluorescent signal.
