Methods: The suppressive effects of BIT on allergic inflammation were investigated using interleukin (IL)-4 mRNA expression, IL-4, IL-6 cytokines, and nitric oxide (NO) production in lipopolysaccharides (LPS)-stimulated RAW264.7 macrophage cells. IL-6 mRNA expression and inflammatory proteins such as cyclooxygenase (COX)-2, nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB), AKT, and mitogen-activated protein kinases (MAPK) were also evaluated in phorbol myristate acetate (PMA) and A23187-stimulated HMC-1 mast cells.
Results: BIT inhibited IL-6 mRNA expression and IL-4, IL-6 cytokines in LPS-stimulated RAW264.7 cells. BIT also decreased NO production in LPS-stimulated RAW264.7 cells. In addition, BIT reduced IL-6 mRNA expressions in the PMA and A23187-stimulated HMC-1 cells, and also regulated AKT, NF-κB, and the MAPK pathway via inhibiting the p-NFκB, p-AKT, p-Erk and p-p38 proteins in HMC-1 cells.
