In this study, the aqueous extracts of licorice, containing flavonoid glycosides, were cultured with Grifola frondosa mycelia to produce β-glucosidase. The biological acitivities of fermented licorice extract were then studied. β-glucosidase converted the glycosides, liquritin and isoliquiritin, into the aglycones, liquiritigenin and isoliquitigenin. A licorice extract fermented with G. frondosa mycelia (FLEx) had antioxidant and anti-inflammatory activities. The photoprotective potential of FLEx was tested in human dermal fibroblasts (HDFs) exposed to ultra-violet A (UVA). FLEx was found to have an inhibitory effect on human interstitial collagenase (matrix metalloproteinase-1, MMP-1) expression in UVA-irradiated HDFs. The liquiritigenin produced from FLEx decreased the expression of MMP-1 by approximately 28%, while isoliquiritigenin had negligible effects. The treatment of UVA-irradiated HDFs with FLEx resulted in a dose-dependent decrease in the expression level of MMP-1 mRNA. The expression of MMP-1 mRNA in UVA-irradiated HDFs was significantly reduced by liquiritigenin at a concentration of 5 µg/mL, while the MMP-1 mRNA level in isoliquiritigenin-treated HDFs remained constant. FLEx also stimulated collagen biosynthetic activity in fibroblasts. Cellular senescence leads to an increase in senescence-associated β-galactosidase (SA-β-gal) activity, which can be used as a biomarker to identify senescent cells. Treatment with FLEx resulted in a dose-dependent, statistically significant decreased in SA-β-gal activity, indicating reduced senescence. The safety and efficacy of the 3% FLEx cream, formulated as an anti-wrinkle agent, were then investigated. Compared to the placebo, dermatologist and instrumental evaluation showed that the anti-wrinkle cream resulted in a statistically significant improvement in wrinkles 12 weeks after application. Evaluation of skin safety indicated non-irritating properties. These results indicate that the formulated FLEx product is an effective anti-wrinkle cream.
