These amides have known to be anticancer, antibacterial, antifungal, antithrombotic and anti-inflammatory, and previous studies have supported their potentials for industrial use. Accordingly, this study was undertaken for examining the possibility of TSD-201 as active ingredient for cosmetic raw material, functional food supplement, and new drug through a series of experiments. Melanin biosynthesis, which is a series of processes that biosynthesize eumelanin and pheomelanin, determines skin color. It occurs in melanocytes in animals and is known to be closely related to MC1R signaling. Decreasing tyrosinase activity through inhibition of MC1R signaling controls melanin synthesis, whitening skins. Kojic acid and arbutin, which are currently used as the active ingredients of cosmetics for skin whitening, have been known to be inhibitors of this signaling process. In this study, the inhibitory effects of melanin formation with curcumin, dimethoxycurcumin, bisdemethoxycurcumin, gedunin, and TSD-201 were examined using B16F10 mouse melanocytes and zebrafish embryos. Among them, inhibition of tyrosinase activity by gedunin and TSD-201 was observed in the B16F10 cell line and showed a significant decrease in melanin formation. This phenomenon was also found in zebrafish embryos. In addition, as a result of confirming the inhibitory mechanism of these two compounds, the expression level of the genes related to melanin biosynthesis as Mitf, Tyr, and Trp-1 was measured and confirmed that these two natural compounds inhibited the melanin production by controlling the expression level of the genes. Biliary atresia is the most common congenital disease and genetic disorders are considered as the cause of the disease, but the exact reason is not known yet. In patients with biliary atresia, biliary ducts are narrowed or blocked, and thus bile acids do not secrete into the small intestine, referring accumulation of bile acids in the liver. Finally, the accumulated bile acids activate autoimmune system. There is no available medicine to treat this disease at the present time. Kasai surgery is the only solution to minimize the disease, and there is no other option than prescribing UDCA, before and after the surgery to relieve symptoms. Therefore, it is urgent to develop drugs that can reduce the amount of bile acids in the liver, postponing the development of cirrhosis. In this study, a human hepatocarcinoma cell line HepG2, a mouse hepatocyte cell line AML12, and mouse primary hepatocyte cells were used to examine the level of gene expression in relation to bile acid biosynthesis and pro-inflammatory cytokines after treatments of piperine, TSD-201, and two derivatives. As a result, the expression of bile acid production-related genes was not changed or increased with the treatments of two analogues and piperine. TSD-201 alone decreased expression levels of CYP7A1 and CYP27A1. In addition, it was confirmed that the amount of bile acid with the treatment of TSD-201 was significantly reduced in the HepG2 cell. Performing the western blot confirmed that the expression level of CYP7A1 was reduced due to the decrease in the amount of HNF-4α protein, acting as a transcription factor that regulates the expression levels of CYP7A1 and CYP27A1. In addition to this finding, TSD-201 inhibited TLR4 signaling process induced by LPS in human and mouse hepatocytes, and it strongly inhibited the generation of inflammation, which is a result of excessive immune response. This study confirmed that TSD-201 could play an important role as a drug to significantly improve patients with biliary atresia before and after Kasai surgery. Altogether, we developed TSD-201, a natural product derived from P. longum, presumably used in industries as an active ingredient possessing anti-melanogenic property, strong inhibitory effect on bile acid production, and anti-inflammatory effect in liver. This demonstates that TSD-201 could not only replace cosmetic ingredients with the currently used whitening chemicals, but also could be used as a drug to prevent severe damages in liver cells and tissues before and after Kasai surgery in patients with the rare disease, biliary atresia.
