AD 병인에는 두 가지 가설이 있다; 콜린성 및 아밀로이드 가설. 본 연구는 뽕나무 뿌리 껍질의 n- 부탄올 (n-BuOH) 분획물의 생리활성 물질 분리에 목적을 두었다. 총 페놀 성분 (TPC) 및 총 플라보노이드 성분 (TFC) 분석을 통해 피토케미칼 특성을 분석했다. 분획물과 화합물의 항산화 활성을 DPPH, ABTS 라디칼, 및 ONOO- 소거 분석을 통해 확인했다. 또한, acetylcholinesterase(AChE) 및 butyrylcholinestearse(BChE) 의 억제 분석을 통해 항알츠하이머 활성을 확인했다. 뽕나무 뿌리껍질은 메탄올로 환류 추출하여 동일한 부피 (1:1)의 증류수 (H2O)를 가하여 순차적으로 분획하여 CH2Cl2, EtOAc 및 n-BuOH 분획물을 수득 하였다. n-BuOH 분획물은 다이아이온 칼럼 크로마토 그래피를 수행했고, 0 %, 40 %, 60 % 및 100 % 메탄올 분획을 수득 하였다. 활성 유도 선량분획법을 통해 화합물 1-5를 분리했다; mulberroside A (1), mulberroside B (2), kelampayoside A (3), esculin (4) 및 xeroboside (5). 분리한 화합물 중에서 화합물 3-5이 이 연구에서 처음으로 보고되었다. 화합물의 구조는 1H 및 13C NMR의 분광학적 방법과 문헌 치와 비교하여 규명되었다. 분리 화합물 대부분은 라디칼 소거와 ONOO- 억제 활성을 나타냈고 화합물 1에서 강한 활성을 보이며 이어서 화합물 2 가 뒤 따랐다. 효소 동역학 분석을 통해 화합물 1-4는 AChE에 대해 혼합형 억제로 확인되었고 화합물 5는 비경쟁적 억제로 확인되었다. 또한 BChE 억제의 경우, 화합물 5는 혼합형 억제로 확인되었다. 이를 토대로 본 연구는 뽕나무 뿌리껍질의 극성 분획물에서 분리한 화합물이 산화스트레스와 알츠하이머병의 잠재적인 천연 치료제로 개발 될 수 있다고 시사한다.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder which leads to the loss of memory and behavioral changes. It has been one of the leading global health burdens being the fourth leading cause of death which is prevalent in the Europe and USA. The present study is focused on the isolation of bioactive substances from the n-butanol (n-BuOH) fraction of Morus alba L. root bark extract. Phytochemical screening assays like total phenolic content (TPC) and total flavonoid content (TFC) were analyzed. Anti-oxidant potential of the fractions and isolated compounds were evaluated by using the 1,1-diphenyl-2-picryl hydrazyl (DPPH), 2ʹ-azinobis (3-ethylbenzthiazoline-6-sulphonic acid) diammonium salt (ABTS) and peroxynitrite (ONOO-) scavenging assays. In addition, the anti-AD potential of the subfractions and compounds were scrutinized by using the acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibitory assays. The root bark of Morus alba L. was extracted with methanol (MeOH) under reflux and subjected to the equal volume (1:1) of distilled water (H2O) which was partitioned sequentially to obtain methylene chloride (CH2Cl2), ethyl acetate (EtOAc) and n-BuOH fractions. Further, Diaion column chromatography was performed with the n-BuOH fractions to obtain 0%, 40%, 60% and 100% MeOH subfractions. Bioactivity guided fractionation led to the isolation of the compounds 1-5; mulberroside A (1), mulberroside B (2), kelampayoside A (3), esculin (4) and xeroboside (5), among them compounds 3-5 have been reported for the first time in this study. Structure elucidation of the compounds was performed by using 1H and 13C NMR along with the published spectral data. Most of the isolated compounds revealed the active radical scavenging and ONOO- inhibitory potential with the strong activity in compound 1 which was followed by compound 2. From the enzyme kinetic analysis, compounds 1-4 showed the mixed type of inhibition for AChE except compound 5 showing the noncompetitive type of inhibition mode. Moreover, in the BChE inhibition kinetic assay, compound 5 showed the mixed type of inhibition mode. Taken together, the present study suggests that the compounds isolated from the polar fraction of Morus alba L. root bark extract can be developed as a potential natural therapeutic agent against the oxidative stress and AD.
