Acetylcholinesterase (AChE) 및 butyrylcholinesterase (BChE)에 의한 아세틸 콜린의 과도한 분해는 기저 전뇌의 콜린성 뉴런의 퇴행과 AD를 초래한다. 활성 산소 종은 산화스트레스의 주요원인으로 알츠하이머병의 잠재적인 원인인 신경세포 사멸을 유발한다. 뽕나무는 뽕나무과에 속하는 치료상 중요한 식물이다.“상백피”라고 불리는 뽕나무 뿌리 껍질은 중국 전통 의약으로 당뇨, 염증, 고혈압 치료에 쓰여왔다. 뽕나무는 isoprenylated flavonoids, Diels–Alder type adducts, benzofurans 같은 페놀성 화합물의 풍부한 공급원이다. 상백피의 성분은 다양한 생물활성으로 인해 연구자들에게 큰 관심을 끌고 있다. 본 연구의 목적은 sanggenofuran A (1), betulinic acid (2), cathafuran A (3), mulberrofuran D (4) and sanggenol L (5)을 포함하는 상백피의 비극성 분획물에서 분리한 화합물의 cholinesterase 억제 활성과 ONOO- 소거 활성을 확인하는 것이다. 화합물은 반복적인 silica gel, RP-18 gel, sephadex LH-20 칼럼 크로마토 그래피를 사용하여 활성 유도 선량분획법에 따라 분리되었다. 화합물을 1H, 13C NMR, HMBC, COSY, NOESY를 포함한 분광학적 방법과 문헌치와 비교하여 확인하였다. 분리된 화합물의 항 알츠하이머 활성은 AChE 및 BChE 억제 분석을 통해 평가하였다. 또한 ONOO- 소거 활성을 통해 화합물의 항산화 활성을 평가하였다. 결과적으로 모든 화합물은 강력한 ONOO- 소거 활성과 함께 강력한 AChE및 BChE 억제 활성을 나타냈다. 특히 화합물 3은 양성 대조군인 berberine과 비교하여 가장 강력한 AChE (IC50 값 14.9 ± 3.54 μM) 및 BChE (12.6 ± 1.63 μM) 억제 활성을 나타냈다. 또한 강력한 ONOO- 소거 활성 (2.85 ± 0.01 μM) 을 나타냈다. 이 결과는 이 연구에서 처음 보고되었다. 또한, 효소 동역학 연구에서, 모든 화합물은 AChE 및 BChE에 대해 혼합형 억제를 나타냈다. 시험된 화합물 중에서, 화합물 1, 3, 4는 prenyl과 또는 geranyl 기를 가지는 arylbenzofuran이고 화합물 2는 triterpenoid, 화합물 5는 isoprenylated flavonoid이다. 우리의 결과는 prenyl 또는 geranyl 기를 가지는 arylbenzofuran및 flavonoid 또는 triterpenoid 골격이 cholinesterase 억제 활성에서 유망한 활성을 보인다는 것을 나타냈다. 따라서, 상백피로부터 분리 된 화합물은 cholinesterase의 억제를 통해 알츠하이머 병 관리에 중요한 역할을 할 수 있다.

Alzheimer’s disease (AD) is one of the serious neurological disorders which is prevalent among elderly people, especially above 65 years. Degeneration of cholinergic neurons in the basal forebrain due to excessive breakdown of acetylcholine by acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) leads to AD. Reactive oxygen species is a leading cause of oxidative stress, causing neuronal cell death which is also a potential reason for AD. Morus alba L. is a therapeutically important mulberry plant, belonging to the Moraceae family. The root bark of Morus alba has been used for long in traditional Chinese medicine as “Sang-Bai-Pi” for the treatment of diabetes, inflammation and hypertension. Morus alba root bark is a rich source of natural isoprenoid substituted phenolic compounds such as flavonoids, Diels–Alder type adducts, and benzofurans. Nowadays, constituents from Morus alba root bark are of great interest to researchers due to its diverse bioactivities. The aim of the present study was to determine the anti-AD effect via cholinesterases (ChEs) inhibition and peroxynitrite (ONOO-) scavenging activity of the compounds isolated from the non-polar fraction (CH2Cl2) of the Morus alba root bark including, sanggenofuran A (1), betulinic acid (2), cathafuran A (3), mulberrofuran D (4) and sanggenol L (5). The compounds were isolated according to the bioactivity-guided fractionation using repeated silica gel, RP-18 gel and sephadex LH-20 column chromatography. The compounds were identified by spectroscopic methods, including 1H NMR, 13C NMR, HMBC, COSY and NOESY and compared with the published data. The anti-AD effect of the isolated compounds was evaluated by AChE and BChE inhibitory assays. ONOO- scavenging activity was performed to analyze the antioxidant activity of the compounds. As a result, all compounds exhibited strong AChE and BChE inhibitory activities along with potent ONOO- scavenging activity. However, compound 3 demonstrated the most potent AChE (IC50 value 14.9 ± 3.54 μM) and BChE (12.6 ± 1.63 μM) inhibitory activities as compared to berberine, positive control used in the assay. It also showed strong ONOO- scavenging activity (2.85 ± 0.01 μM). This result has been reported for the first time in this study. Moreover, in the enzyme kinetic study, all compounds revealed mixed type inhibition mode against AChE and BChE. The tested compounds were 2-arylbenzofurans having prenyl or geranyl groups except (4), an isoprenylated flavonoid and (5), a triterpenoid. The results suggested that the presence of prenyl or geranyl group on 2-arylbenzofurans and flavonoid, or a triterpenoid skeleton could be promising targets in ChEs inhibitory activity. Thus, the compounds isolated from the Morus alba root bark may play a significant role for the management of AD via ChEs inhibition.
